Identification and Molecular Characterization of an Avian Infectious Bronchitis Virus Strain HN104

Abstract:

A virus was isolated from a suspected of suffering from avian infectious bronchitis chicken farms in Henan province in April 2010.The virus was confirmed as a genetic variation occurred avian infectious bronchitis virus by SPF chicken embryos induced dwarf experiment, chicken red blood cell agglutination test, interference with Newcastle disease virus proliferation assay and RT-PCR tests. Its allantoic fluid hemagglutination titer which for chicken red blood cells is 0.However,viruses’ HA titer is 27 after treatment by trypsin.EID50 (50% egg infectious dose) is 10-5.5/0.1 mL; It can significantly interfere with Newcastle Disease Virus La Sota strain proliferation in the chick embryo; SPF chickens infection Experiments showed that the virus can cause swelling of the kidney and spotted kidney. The S1 cDNA was 1617 bp. Phylogenetic tree showed that the kinship between IBV Massachusetts strain, T strain and 4/91 strain is distantly related less than 78%.
