Effect study of Mycoplasma suis on the maturation of Dendritic Cells

Abstract:

To observe the phenotype and function changes of the dendritic cell (DC) 2.4 in the initial study of Mycoplasma suis (M. suis) on DC maturation. DC 2.4 was proliferated by cell culture. DC2.4 were divided into five groups: blank control group: there was no treatment; experimental group: M. suis 0.1, 1.0, 10 μL/mL groups, the purified M. suis with the corresponding dose were added in the culture medium; positive control group: lipopolysaccharide with final concentration of 1 μg/mL was added into the culture medium. There were 6 holes in each group. Expression of DC2.4 cell surface CD80, CD86, MHC-Ⅱ molecule in dendritic cells cultured for 24 hours were detected by flow cytometry, followed by parallel in vitro mixture with lymphocyte reaction. Types of DC on T cells with stimulation capacity were observed by MTT. The results of flow cytometry showed that, M. suis significantly inhibited DC2.4 surface molecules (CD80, CD86 and MHC-Ⅱ) expression (P<0.01), lower than which contracted with positive control group. In addition, M. suis significantly inhibited the dendritic cells to T cell activation capacity, and there were significant differences between positive control group and blank control group (P<0.01) in the mixed lymphocyte reaction. M. suis could inhibited the activation of dendritic cells mature, and the nagative regulatory role of the mature DC in immune function may provide research data for M. suis immunology study.
