Establishment of Tissue Culture and Rapid Propagation System for Rare Medicinal Bletilla striata

Abstract:

To establish the system of tissue culture for Bletilla striata, an experiment was conducted to screen the concentrations of sucrose, exogenous hormones (6-BA, IBA, IAA and NAA) and activated carbon (AC), light duration and hardening-seedling matrix for seed germination and growth taking mature seeds as original explants. The results showed that the seeds germinated after 7 d in the medium MS+4 g/L agar+20 g/L sucrose+ light (200 lx) 24 h/d; the number of cotyledon reached 5 with supplemental 1.5 mg/L 6-BA+0.5 mg/L NAA+1.0 g/L AC; the root number and length respectively reached 3 and 4 cm with supplemental 1.5 mg/L IBA+1.0 g/L AC; and the seedling survival rate reached 100% under the matrix vermiculite: peat (1:2). These findings would provide a theoretical basis and technical support for large-scale cultivation, pharmaceutical production and wild resource protection of Bletilla striata.
