Development of Histone Methylation and Demethylation

Abstract: In eukaryotic genes,histones are subject to covalent modifications and histone lysine methylation plays an importance in epigenetics; the different methylation sites have different mechanisms. Histone H3K9 methylation is involved in heterochromatin formation,gene silencing; H3K4 methylation contributes to transcriptional activation and K36 methylation is linked to RNAPⅡ.The first report shows histone methylation is reversible and discovered lysine-specific histone demethylases:LSD1 and Jmjc-domain-containing histone demethylases. They can demethylate different levels of histone H3K4, H3K9and H3K36 methylation, are associated with gene transcription.
