Polymorphisms and Bioinformatics Analysis of the Goat Growth Hormone Gene in the 5′flanking Sequence

Abstract: The polymorphisms of two sites of GH gene in the 5′ flanking region from 168 Huanghuai goats、Changjiangsanjiaozhou white goats and Boer goats were analyzed by PCR-SSCP, At the same time, the three communities were analyzed to check whether they were in balance. Results: both of the two sites of GH gene were polymorphic, there were six genotypes and two mutations with the first pair primers, three genotypes and three mutations with the second pair primers. communities analysis shows that all the three breeds in the first site were in imbalance, which suggests that artificial selection might be possible in this site; and all the three breeds in the second site were in balance. Furthermore, GH gene in the 5′ flanking sequence were analyzed by bioinformatics software assay, which shows that transcription factor combining sites with high score, such as IA-1、PIT-1、Gsh-2、C/EBP et al, exist in the polymorphic sites, and the mutation of T→C(264) results in the disappearance of PIT-1 and Gsh-2 combining sites, Further studies need to be carried out to check such sites will affect the expression of GH gene, and the growth performance of goat.
