  Cloning and Sequence Analysis of Genetic Variation on NS2-3 of Bovine Viral Diarrhea Virus HB-DCZ Strain Zhao Yuelan, Guo Hongbin, Zhang Lei, Qin Jianhua, Zhang Ning, Zhang Baoning 2007, 23 (6):  1-1.  Abstract + ( 1613 )   【Objective】Bovine Viral Diarrhoea-Mucosal Disease Virus HB strain was a field strain isolated from a dairy cows flock in HeBei province . The study were progressed in order to realizing characters of HB strain, providing theory base for illuminating pathogeny, controlling infection and prevalence of BVDV;【Method】Genetic Variation on NS2-3 of Bovine viral diarrhea Virus HB-DCZ strain were amplified by RT-PCR. The product of PCR was cloned into pMD18-T vector, and then transfected into JM109. The recombinant plasmids were extracted and amplified by PCR and identificated by EcoRⅠand Hind Ⅲ enzyme digestion. The nucleotide sequence of the isolated virus NS2-3 was sequenced and amino acid sequence was inferred. Sequence analysis was performed with the aid of DNAstar softerware；【Results】The result showed that the obtained fragment was approximate 665bp by RT-PCR. There was no insertion in the isolated virus genome. EcoRⅠand Hind Ⅲ digestion of the recombinant plasmids resulted in two bands with sizes 702bp and 2635bp, indicating the specificity of the PCR. The cloned NS2-3 segment of HB-DCZ strain contains 665bp nucleotides and 208 amino acid. The homologies of nucleotide sequence of NS2-3 gene of HB-DCZ strain with other BVDV strains were 99.1%(184)，97.4% (ZM195), 92.3% (Osloss), 77% (OregonC24V ), 76.4% (NADL), respectively. The HB-DZC virus strain had no exogenous sequence insertion, gene recombination, gene rearrangement or gene deficiency. However, some nucleotide sequences were replaced. 【Conclusion】The HB-DZC was closed related to BVDV strains Osloss, 184, ZM195 nor the strains C24V, NADL; All the above results proved this virus was BVDV and which belongs to subtype Ib.

  Effect of Colloidal Gold on immunological function in mice Tian Ye,, Cui Yanshun, Lou Hua,, Li Jianliang,Yan Pingping 2007, 23 (6):  7-7.  Abstract + ( 1594 )   The experiment designed twelve experimental groups to inject laboratory mice ,including four kinds of diameters colloid gold ,each of it have three kinds of doses. This experimentation adopt MTT ,RIA , LDH assays to mensurate the activity of lymphocyte proliferating and abdominal cavity macrophage, content of IL-2 in serum and induction in spleen lymphocyte and activity of NK cell. The results shows that different effects on mice which injects CG for mice.0.3 mL 0.2 mL of 15 nm , 10 nmCG can enhance the activity of lymphocyte proliferating and increase content of IL-2 in serum and induction in spleen lymphocyte, meanwhile they can also enhance the activity of abdominal cavity macrophage and NK cell notably. 0.1 mL doses can improve also but unconspicuous. The effects on eath doses of 5nm and 20nmCG is fluctuate.they can improve the four criterions but unconspicuous, the low doses have no effect. We can learn that the middle and high doses of 15 nm and 10 nm colloidal gold can enhancing specific and nonspecific immune response in laboratory mice.

  Effect of Wilting on Chemical and Physical Characteristics of Silage with Corn Stalk Yu Ruhua, Mo Fang, Zhao Lihua, Zhang Xiaoming, Chen Yao 2007, 23 (6):  13-13.  Abstract + ( 1521 )   【Objective】The influence of wilting on nutritive composition was determined for four different silage of corn stalk.【Method】The silage were ensiled at the first, third, fifth, and seventh day after harvested.【Results】Although the pH and ammonia concentrations of silage were higher for ensiled at the seventh day than firs day, lactic acid, CP, NDF, and ADF content of silage had no consistent effect.【Conclusion】Results of trials indicate that the corn stalk could be ensiling among five days after harvested.

  Mechanism of Keratin Degradation by Streptomycete sp.B221 Zhu Xiaofei，Zhang Ling，Zhao Pingzhi，Wang Ruiyong 2007, 23 (6):  18-18.  Abstract + ( 1763 )   Streptomycete sp. B221 was cultured on chicken feather in a mineral solution. The degrading rate reached maxium from 2nd day to 3rd day and reached as much as 82.7% at 5th day. Keratinase activity was detected in the supernatant of the medium and the largest enzyme activity occurred at 4th day. The incomplete correspondence between the keratin degrading rate and keratinase activity revealed the existence of non-enzymatic degradation pathway of keratin. The sulfur in the keratin was mainly removed into the medium in the form of sulfate. Sulfite was also found in the medium. The concentration of sulfite changed relating to keratinase activity, keratin degrading rate, soluable protein and thiol concentration, which indicates sulfite plays important role in the process of keratin degradation. Both enzymatic pathway (degradation by keratinase) and non-enzymatic pathway that is sulfitolysis occurred in the degradation process by streptomycete sp.B221.

  Study on the lZP3 mRNA Expression after siRNA Interference Zhuang Shuzhen, Guo Jina, Zheng Yaohu, Li Jingjing, Zhang Fuchun 2007, 23 (6):  23-23.  Abstract + ( 1460 )   【Objective】In order to explore lZP3 gene in the oocytes and sperms combination mechanism and control the population of the Lagurus lagurus.【Method】The RNA interference vector pGenesil-ZP31 and the lZP3 expression vector were co-injected into the mice by Hydrodynamics-based transfection method (HD).【Results】 Results of semi-quantative RT-PCR and real-time PCR showed that, the interference effect was occurred between 8 h and 10 d when co-injected the 2 vectors. Study of sequential injections revealed that prior injection of pGenesil-ZP31 produced a significant suppression at 8 h and 1 d, which disappeared after 3 days. However, injection of p-ACLC followed by pGenesil-ZP31 at 1 h, 2 h, 4 h and 8 h, the expression of lZP3 mRNA was also suppression, which disappeared after 24 hours.【Conclusion】Taken together, these results demonstrate that vector-based in vivo RNAi is a time-dependent process and offers the use for references in studying the mechanism of siRNA interference the Lagurus lagurus ZP3 at the oocytes level.

  The Progress of Bovine Spermatogonial Stem Cells Researches Wu Jianying, Hu Tiejun, Yue Zhanpeng, Zhang Xueming, Li Ziyi, Li Dexue, 2007, 23 (6):  28-28.  Abstract + ( 1555 )   Spermatogonial stem cells (SSCs), the postnatal male germline stem cells, reside on the base membrane of the seminiferous tubules in testes. They are not only capable of self-renewing and differentiating but also the only adult stem cells in normal postnatal body that undergo self-renewal throughout life and transmit gene to offspring. According to the related reports most recently, the biological properties separation, purification, cryopreservation, in vitro culture, establishment of immortalized cell line and transplantation of bovine spermatogonial stem cells were discussed.

  Study on the Structural Layer of Crude Fiber in Roughage Li Hua, Kong Xingang, Wu Guolian 2007, 23 (6):  32-32.  Abstract + ( 1220 )   The structural layer of crude fiber in the roughage of cereal was investigated by enzymatic hydrolysis. The crude fiber consists of two layers, i.e., the free type of cellulose and hemicellulose, it was easy to be hydrolyzed by enzyme; and the combined type of cellulose and hemicellulose bonded to lignin, it was hard to be hydrolyzed by enzyme. It was found that the former was hydrolyzed by enzyme containing 80.98% of the total hemicellulose and containing 19.31% of the total cellulose in cotton stalk. The former was hydrolyzed by enzyme containing 60.23% of the total hemicellulose and containing 44.36% of the total cellulose in maize stalk. The former was hydrolyzed by enzyme containing 46.20% of the total hemicellulose and containing 43.04% of the total cellulose in wheat stalk. The former was hydrolyzed by enzyme containing 47.60% of the total hemicellulose and containing 51.80% of the total cellulose in rice stalk. It suggests that this could be the main reason for lower ratio of digestibility and metabolism of roughage.

  Detection on VTEC with Multiplex Fluorescent Polymerase Chain Reaction Bei Feng,, Zhou Shengyang, Wang Chao, Zhang Guiming, Li jianliang, Cui Yanshun 2007, 23 (6):  37-37.  Abstract + ( 1675 )   To develop a nove1 sensitive, specific and lower cost method applicable for detection of VTEC. Two primers were designed respectively based on the gene of VT1 and VT2,and the VTEC gene were detected by PCR which includes SYBR Green I, a double-stranded DNA-specific fluorescent dye, and the sequence specific primers, The results showed that the method possessed high specificity and sensibility for detection of VTEC and achieved 10cfu/g.The method could detect VTEC from clinical specimen and food. The method was quickly，convenient, accurate, high efficient and in favor of early diagnosis and detection of VTEC.

  Polymorphisms and Bioinformatics Analysis of the Goat Growth Hormone Gene in the 5′flanking Sequence Li Zhongyuan, Wang Feng, Pang Xunsheng, Wang Ruifang, Wang Ziyu 2007, 23 (6):  42-42.  Abstract + ( 1473 )   The polymorphisms of two sites of GH gene in the 5′ flanking region from 168 Huanghuai goats、Changjiangsanjiaozhou white goats and Boer goats were analyzed by PCR-SSCP, At the same time, the three communities were analyzed to check whether they were in balance. Results: both of the two sites of GH gene were polymorphic, there were six genotypes and two mutations with the first pair primers, three genotypes and three mutations with the second pair primers. communities analysis shows that all the three breeds in the first site were in imbalance, which suggests that artificial selection might be possible in this site; and all the three breeds in the second site were in balance. Furthermore, GH gene in the 5′ flanking sequence were analyzed by bioinformatics software assay, which shows that transcription factor combining sites with high score, such as IA-1、PIT-1、Gsh-2、C/EBP et al, exist in the polymorphic sites, and the mutation of T→C(264) results in the disappearance of PIT-1 and Gsh-2 combining sites, Further studies need to be carried out to check such sites will affect the expression of GH gene, and the growth performance of goat.

  Effects of Different Levels of Zinc on Semen Quality and Blood Biochemical Parameters in Holstein Bulls Xin Fuyu,, LI Wenli, Hou Minghai, LI Yanqin, Wang Lingling, Song Jie,Zhong Jifeng 2007, 23 (6):  47-47.  Abstract + ( 1721 )   【Objective】Thirty Holstein bulls assigned randomly to five groups was conducted to study the effect of different levels of zinc on semen quality and blood biochemical parameters.【Method】This experiment involved 5 dietary treatments, consisting of one control group fed basal diet and four levels of zinc which is 50,80,110,200mg/kg. The experimental period lasted 8 months. 【Results】The results shows that: (1) The effect of different levels of zinc on the sperm motility, ejaculate volume and the sperm concentration was extremely significant(P<0.01); but that on the acrosomal integrity of sperm was not significant(P>0.05); and zinc supplemental could reduce the percentage of abnormalities significantly(P<0.05); (2) The effect of different levels of zinc on the testosterone contents in serum was extremely significant (P<0.01); but that on serum zinc, zinc and testosterone contents in seminal plasma was not significant (P>0.05); (3)The effect of different levels of zinc on the activity of GOT in seminal plasma was extremely significant (P<0.01), but that on LDH was not significant(P>0.05) and which on AKP,GPT was significant(P<0.05); The effect on the activity of GPT、GOT in serum was extremely significant (P<0.01),but that on AKP、LDH was not significant (P>0.05). 【Conclusion】All of the variety of physiological and biochemical parameters analysed in each group indicated that the optimal supplementation of zinc was 110mg/kg by the dry matter in this experiment.

  Regeneration plants of cotyledon,hypocotyl and root of Xinjiang Big Leaf Alfalfa(Medicago.sativa.L) Pan Jingli,, Zeng Youling, Zhang Fuchun 2007, 23 (6):  52-52.  Abstract + ( 1806 )   Following the development of plants resistance and transgenic techonology, breed of new resistant alfalfa by transforming heterogenous gene has being become more and more remarkable to pepole.So,establishing a high-frequence system of regeneration plants is the base of high efficiency transform for heterogenous gene. Cotyledon, hypocotyl and root of 5-7 day-old seedling of Xinjiang big leaf alfalfa were selected as explants. MS and 8 groups of 2,4-D with different concentrations from 0.1mg/L to 3mg/L were used to induce callus, then MS and 2,4-D2mg/L and 10 groups of KT with different concertrations from 0.01mg/L to 0.5mg/L were used to induce callus; the shoots were induced in the medium of MS with additional 6-BA 0.5mg/L and NAA 0.05mg/L; MS was used to produce roots. Results showed that: callus tissues had much better growing status and could be used to produce differentiation shoots in medium of MS with 2,4-D 2.0mg/L and KT 0.2mg/L, the averaging calli frequences were 93.1%, 100% and 100% for cotyledon, hypocotyl and root; After growing 40-80 days in the medium of inducing regeneration shoot, callus tissues which originated from cotyledon, hypocotyl and root produce regeneration shoots and its averaging regeneration shoots frequence was 50%, 78% and 50% respectively; when shoots grew to about 2cm height, it was transferred into medium of MS to root; afer 14 days, regeneration plants were transferred into flower-soil and its survivable rate was more than 90%. All of cotyledon, hypocotyl and root can obtain regeneration plants in this culturing system. Moreover root can act as a good material of explant to obtain regeneration plant while researches in this field are still poor.

  The mtDNA D-loop Polymorphism and Origin of 4 Estern Asia Black Goat Populations Hou Lei, Wang Jianmin, Li Peipei, Li Jue, Xing Feng 2007, 23 (6):  57-57.  Abstract + ( 1711 )   In order to investigate the origin and differentiation of indigenous goat populations, clarify the genetic background to provide the sound utilization with basic information, partial sequence of mtDNA D-loop of 39 black goats from 3 Chinese and 1 Korean population were sequenced and analyzed. 441 bps sequence was analyzed, 45 polymorphic sites were defined, including 11 singlton polymohic sites and 44 parsimony polymorphic sites, all the 55 sites defined 29 haplotypes, and haplotype diversity is 0.984. Phylogenetic tree defined 3 major lineages, C.aegagrus clustered with lineage A. The relative relationship among populations was also analyzed through pairwise distribution and genetic distance between haplotyoes. The results show that the 4 black goat populations are abundant in genetic diversity, at least have 3 different maternal origins, C.aegagrus is one of the origins of domestic goat.

  Establishment and application of colloidal gold immunochromatographic assay for quick diagnoise of hog choleras Wang Huijie, Ning Yuchang 2007, 23 (6):  61-61.  Abstract + ( 1507 )   A simple method was developed for the detection of Hog Cholera Virus (HCV) in clinical samples using colloidal gold immunochromatographic assay, and we can discriminate violent and weak virus by using this method. Protein E2 of Hog Cholera Virus that was expressed in E.coli acted as attenuated HCV antigen and the violent HCV that was cultivated and purified by using PK15 cells acted as violent HCV antigen. By using these materials, we developed the immunochromatographic assay test paper for detecting HCV. The test paper still had good stability after it was destroyed by the temperature of 37℃, the sensitivity of the test paper was better in certain degree than the American IDEXX enzyme linked immunity reagent and the indirect agglutination through comparison; experimented with several kinds of masculine blood serums of virus and disease germs, and it demonstrated that the test paper had better specificity. The development of this test paper provides a more practical method for the fast diagnosis of HCV.

  Definition of Typical Body Traits of Duck Race in Fujian Zhang Li, Wang Chang kang, Shao Liang Ping, Liu Hao 2007, 23 (6):  65-65.  Abstract + ( 1542 )   The purpose of this study was to determine the representative body type traits of four Fujian native breeds of ducks, which are Putian Black Duck, Liancheng White Duck, Jinding Duck and Fujian Muscovy. With correlation and cluster analysis, eleven body type traits of the ducks were studied. According to the result of cluster analysis and , some representative traits were picked out from various body type traits. Putian Black Duck can be divided into three types and the representative traits were weight, chest width, and neck length. Liancheng White Duck can be divided into four types and the representative traits were weight, chest width, mouth width, and pelvis width. Jinding Duck can be divided into four types and the representative traits were weight, half-diving length, pelvis length and chest depth. Fujian Muscovy can be divided into five types and the representative traits were weight, body length, chest depth, mouth length and mouth width. The various body type traits can be summed up as just a few indices with cluster analysis, the indices which provide the scientific basis for breeding and improving the native breeds.

  Acute Mammitis of Milk Cow Treatment Contrast Experiment Yang Qianfeng 2007, 23 (6):  69-69.  Abstract + ( 1420 )   Seventy-five milk cows that cause acute mammitis were divided into three groups randomly and different therapeutic methods were adopted. Result is indicated: efficiency and the curative ratio reaches 100% equally if we mixed traditional Chinese medicine and western medicine. Using penicillin and streptomycin to treat, the efficiency is 92% and the curative ratio is 88%. Efficiency is 84% and curative ratio is 80% if we only use traditional Chinese medicine. Penicillin and streptomycin mixing traditional Chinese medicine has complementary advantages and the cure effect is the best. It is worth to apply this method to cure acute mammitis of milk cow.

  Molecular cloning and sequence analysis of porcine musclin Wang Weijie, Guo Yujie, Li Weihua , Lin Maowang, Zhao Lei ,Yang Guoyu 2007, 23 (6):  72-72.  Abstract + ( 1654 )   【Objective】To clone and analysis the porcine musclin.【Method】Total RNA was extracted from muscle tissue and mRNA sequence of gene were amplified by RT-PCR using two designed primers. The PCR products were ligated into the pMD-19Tvector, and then transformed into competent cells of E. coli DH5α.The sequence was analyzed to identify the recombinant plasmid.【Results】Identity analysis showed that the musclin nucleotide sequence shared 86%, 78% and 75% homology with that of human , rat and mouse. The predicted peptide contained a “KKKR” motif and the region homologous to mouse ANP, BNP, and CNP.【Conclusion】The porcine partial musclin gene was successfully cloned in present study and the sequence has been submitted to GenBank(Accession.EF369511).

  Identification of type 2 of swine streptococcus suis from piglets by PCR in Hebei Fan Jinghui, Zuo Yuzhu 2007, 23 (6):  75-75.  Abstract + ( 1375 )   A strain of pathogenic swine streptococcus was isolated and identified from dead piglets in a case of a pig farm. The identification was achieved by phenotypic, biochemical specificity experiment and antimicrobial sensitivity analysis. The results indicate that the pathogen is treptococcus. The colonies of streptococcus suis were purified and liquid-cultured, then the DNA was extracted and objective fragments. The results indicated the strain was swine streptococcus suis type 2.

  The System Establishment of AFLP Technique in Sugarcane Que Youxiong, Lin Jianwei, Chen Tiansheng, Xu Liping, Zhang Muqing, Chen Rukai 2007, 23 (6):  78-78.  Abstract + ( 1580 )   【OBJECTIVE】 In order to establish the AFLP technique system in sugarcane; 【METHOD】 the hybrid parents CP84-1198 and Ke5 and correspondingly the resistance pool and susceptible pool were taken as plant materials and the AFLP(amplified fragment length polymorphism)-sliver stained experiment system was constructed accordingly. Moreover, the due system was used to analyze the drought-resistance character using the cDNA from E.arundinaceus as materials and a DD (differentially display) analysis method, which can also be named as cDNA-AFLP, was established. 【RESULTS】 During the experiment, four pairs of primers which can amplify differentially displayed bands between the resistant parent and susceptible parent, were selected. Among them, one pair of primer was found to produce favorable differentially displayed bands between the control and drought-menace E. arundinaceus. At the same time, both a quick method of silver staining of PAGE and a feasible gel conservation method were set up. And the recovery protocol of band on PAGE was also proved to be convenient and efficient. 【CONCLUSION】 In conclusion, the AFLP technique system in sugarcane analysis has been established in this paper.

  Cloning, Expression and Autophosphorylation of 5 Rice Protein Kinases Liu Qian,, Sun Jian, Li LiYun, Liu LiJuan, Xu Zhouda, Wang HaiJiao, Jin MiaoJing, Liu GuoZhen 2007, 23 (6):  83-83.  Abstract + ( 1326 )   It has been demonstrated that protein kinases play important roles in almost all biological process, e.g. growth and development, regulation of expression, stress responses and signal transduction. The finished rice genomic sequence indicated that at least 1500 protein kinases exist in rice genome. In the current study, five rice kinases has been amplified based their full-length cDNA sequences, their corresponding proteins have been expressed and purified in yeast, the accession number are NM-189878, NM-194133, NM-190508, NM-197522 and NM-197571 respectively. NM-194133 has strong autophosphorylation activity. Based on sequence similarity, the deduced functions of these kinases are biosynthesis regulation of carbon metabolism, seed embryogenesis, cell division regulation, response to biotic/abiotic stimulation etc. This work is helpful for in vitro enzyme assay, protein-protein interaction analysis, and provides an efficient system for high throughput cloning and expression of rice genes.

  The Development of Two-dimensional Electrophoresis and its Application in Agricultural Biological Proteomics Liu Weixia, Pan Yinghong 2007, 23 (6):  89-89.  Abstract + ( 1639 )   Two-dimensional electrophoresis is now the key technique in proteome research.In this review,we discuss recent advances in sample preparation, isoelectric focusing，SDS-polyacrylamide gel electrophoresis,and protein detection for two-dimensional electrophoresis technique, and evaluate the difference gel electrophoresis based on two-dimensional electrophoresis briefly.We also describe the application of two-dimensional electrophoresis in agricultural biological proteomics, deliver the prospect of the two-dimensional electrophoresis technique.

  Effects of Transcription Factor ABP9 Over-expression on Plant Growth and Development Meng Hui,, Zhang Xia, Zeng Rizhong, Fan Yunliu, Zhao Jun 2007, 23 (6):  94-94.  Abstract + ( 1703 )   【OBJECTIVE】 In order to analyze the effects on plant growth and development of over-expressing ABP9, an ABA/stress-responsive transcription factor gene from maize, aiming at genetic engineering crops for abiotic stress tolerance. 【METHOD】 An plant expression vector on which the expression of ABP9 is driven by 35S promoter was constructed and transgenic Arabidopsis lines over-expressing ABP9 were obtained. The morphological, developmental, physiological and gene expression changes in transgenic plants comparing to wild type were analyzed at germination, vegetative and reproductive growth stages. 【RESULTS】 The results showed that under normal growth conditions, comparing to that of wild type plants, the growth and development of transgenic plants was delayed at germination, seedling growth and flowering stages. The stomata pores and leaf ABA levels reduced. The expression of genes involved in ABA signaling increased whereas that of ABA biosynthetic genes decreased. All of the effects are positively correlated with the expression levels of ABP9. 【CONCLUSION】 The data indicates that ABP9 over-expression induces stress responses in transgenic plants even under normal growth conditions and retards growth and development. This suggests that the timing and levels of ABP9 expression should be tightly controlled when using it to improve plant stress tolerance.

  Primary Study of Function of A Rice WRKY19 Gene Promoter Hao Zhongna, Wang Haihua, Wu Xuelong, Guo Zejian 2007, 23 (6):  99-99.  Abstract + ( 1662 )   Using transgenic plants of rice WRKY19 gene promoter (OsW19p::GUS) to do this experiment, the inducible expression characteristic of OsW19p in T1 seedlings, detected by GUS fluorometric measurements revealed that: expression level of OsW19p was increased by ABA(100μmol/L),SA(500μmol/L), high temperature(42℃), low temperature (5℃) and wounding treatments; And was repressed by 2,4-D(1 μmol/L), NaCl(200 mmol/L)and PEG4000(25%) treatments; IAA(5 μmol/L), MeJA(100 μmol/L)and ultraviolet treatments can’t influence expression. Expression level of roots of OsW19p was higher than 35s in transgenic plants, but leaves of it was lower one. Furthermore, expression level of clums of transgenic plants was the highest in the flower stage, followed by flowers, sheathes and leaves, the roots showed the lowest expression.

  Transferring Cell Cycle Protein Gene (CycD2) into Wheat via Micro-projectile Bombardment Liu Qi, Wang Xiaoxia, Tang Zongxiang, Ren Zhenglong 2007, 23 (6):  104-104.  Abstract + ( 1309 )   PDF ( 323952KB )   ( 634 ) The present study utilized mature seeds of wheat (Triticum aestivum L.) cultivar mianyang11 as the target material cultured in vitro, and the plasmid PBI121 which contains the cycd2 cell cycle protein gene and selectable marker NptII gene had been transferred into callus initiated from mature embryo of wheat by micro-projectile bombardment. The result showed that some transformation wheat could be refrained effectively and regenerated from the selection medium containing G418 solution. Using this screening method, we have isolated 11 putative T-DNA tagged mutants with alterations in sensitivity in response to iron deficiency. We have cloned one gene, which mutation caused increased sensitivity to Fe deficiency. Identification and characterization the determinants in Fe nutrition of plant will further our understanding on genetic basis of plant in response to nutrient stress. It will provide new insights on molecular improvement of crops.

The 54 SARP primers could amplify 136 polymorphic bands totally with a mean of 2. 5 bands per primer. The 200 single plants of Luliangyou 996 were tested in laboratory by a specific SRAP marker(Me3Em8)whose bands were complement in hybrid combination seeds. The seed purity was 97.50%, which was same to the field purity (97.50% in Hainan). The results of this research suggested that SRAP technique could be used in seed purity identification of two-line hybrid rice.

The nucleotide and amino acid sequence of ALDH were analyzed. The expression of ALDH was researched by Real Time RT-PCR.【Result】ALDH gene segment was obtained which was 675bp and coding 225aa. The compare of nucleotide sequences indicated that it has 86% identity with rice ALDH1a (AB037421), and 85% identity with maize RF2C（AF348413）. By BLASTp Analysis, it shares 87%, 86% and 60% amino acid sequence identity with rice, maize and Arabidopsis thaliana ALDH, respectively, which containing conservation region of aldehyde dehydrogenase families. The data of Real Time RT-PCR indicated that the expression of ALDH was increased first, and then decreased under the inducing situations. As a whole, the responses of ALDH to salt and drought were much higher than to cold.【Conclusion】In this research, ALDH gene segment was obtained, and we researched the expression of ALDH. It established basis on cloning the span of ALDH in Leymus Chinensis.

Suppression subtractive hybridization (SSH) method was used to identify novel genes regulated by cold stressed in sweetpotato. A full length cDNA coding region of S-Adenosyl-L-Methionine synthase (SAMS) which contains 1182bp was isolated by this way combined with RT-PCR technology. Sequence analysis revealed that this genes hared 85.48 % similarity in gene sequence and 93.6 % similarity in protein sequence with the other SAMS gene in plant. The differences of nucleotides sequences were higher than those of amino acids. The phylogenetic tree was constructed based on the alignment nucleotides sequences elucidated the relationship among different plant species at nucleotide and amino acids level, which could proved some help for the utilization of plant germplasms. Fused expression plasmid vector pET-SAMS was constructed with sams and pET32a. A protein about 63KDa was expressed after 3h induced by IPTG at different temperature in BL21.

  Genetic Diversity Analysis of Onion (Allium cepa L.) Four ISSR primers were screened from 31 ISSR primers. The four ISSR primers generated 39 clear bands, which 31 were polymorphic, averaged 7.75 bands each primer pair. The average percentage of polymorphic bands was 79.48 %. The genetic similarity ranged from 0.552 to 0.960, which indicated a rich genetic diversity of onion (Allium cepa L.) germplasm resources. According the result of UPGMA cluster analysis based on ISSR makers, the tested materials could be divided into five groups at the level of D=0.68. First group including 18 germplasm materials is the Yellow sweet Spanish system. Second group including one germplasm material is Bejo Dyatona cultivar. Third group including 3 germplasm materials is the Yellow Globe system. Fourth group including 9 germplasm materials is the Yellow Globe Danvers system. Fifth group including one germplasm material is the Yellow Danvers system. The results indicated that ISSR technique is useful in revealing the genetic diversity and genetic relationship among onion germplasm materials, providing a scientific basis for genetic breeding and patents selection.

However, previous attempts to extract high quality RNA from tomato fruit using published protocols have proven to be unsuccessful, even the use of protocols developed for the extraction of RNA from other fruit tissue. RNA isolation from tomato fruit is difficult due to tomato fruit containing large amounts of polysaccharides, polyphenol which often coprecipitate and contaminate, RNA during the extraction，thereby affecting both the quality and quantity of RNA when using conventional protocols. Now some commercial kits have not been developed directly for the isolation of high quality RNA from tomato fruit. Here four methods were used to isolate functional RNA from tomato fruit, and the best one was screened out. Using this protocol we obtained good RNA yield (401.6--840.2μg/g) from different stage tomato fruit with low levels of polysaccharides, polyphenolic compounds and protein contaminants as determined by A260/A230 and A260/A280 respectively. Furthermore，this RNA was not degraded，as was demonstrated by visualizing the ribosomal RNA of the samples on 1.2％ agarose gel electrophoresis. The RNA is suitable for cDNA-AFLP, which demonstrated that RNA isolation protocol modified in this paper can be used to related molecular researches．

On the basis of the picture, the majority of hybrids showed the close relation to Corylus avellana L. and the distant relation to Corylus heterophylla Fisch. In the experiment 1, fifty ICR mice were assigned randomly to one of five treatments to receive cigarette smoking time 10, 20, 40, 60, or 80 min, observed the mice behaviors. The results showed that 60 min was prior in this experiment. In the experiment 2, one hundred and eighty ICR mice were assigned randomly to one of three treatments with 10 mice per group. The control group orally administered physiological saline, group I orally administered physiological saline and received cigarette smoking, and group II orally administered Pu’er tea extracts and received cigarette smoking. The mice serum was sampled on d 7, 14, 21, 28, 35, or 42, and the serum nicotine and cotinine concentrations were determined. The nicotine concentrations of group I and II were higher (P < 0.01) than those of the control during the period of the second experiment, and the nicotine concentrations of group II were lower than those of group I on d 14 (P = 0.01), 21 (P = 0.02), 28 (P < 0.01), 35 (P < 0.01), or 42 (P < 0.01). The cotinine concentrations were no significant difference between group I and II. The findings of this paper showed that oral administration of Pu’er tea extracts could reduce serum nicotine in passive smoking mice.

The average yield was 2.46%. The quantitative analyses of 21 chemical compounds have been conducted by capillary GC method. The main chemical compounds were: linoleic acid (34.80%), linolenic acid (21.25%), palmitic acid (12.31%), oleic acid (9.20%), linoleic acid methyl ester (4.12%), lauric acid (3.74%), linolenic acid ethyl ester (1.62%) and eugenol (1.22%), etc. The recoveries ranged from 84.02% to 96.58%. Good reproducibility was obtained with RSD ranging from 1.82% to 4.87%. The results of the experiment showed that the analytical method is rapid, sensitive and suitable for analysis of chemical compounds of volatile oil in Rosa Roxburghii Tratt.

One hundred and eighty ICR mice were assigned randomly to one of three treatments with 10 mice per group. The control group orally administered physiological saline, groupⅠorally administered physiological saline and received cigarette smoking, and group Ⅱ orally administered Pu’er tea extracts and received cigarette smoking. After 35 d of this experiment, growth performance and blood parameters were analysized. The final body weight and average daily gain of group Ⅰ mice were lower (P < 0.05) than those of group Ⅱ, and group Ⅱ lower (P < 0.05) control group. The serum hemoglobin and carboxyhemoglobin concentrations of control group and group Ⅱ were no significant difference, but were lower (P < 0.05) than those of group Ⅰ. The hemoglobin concentrations of group Ⅰ and Ⅱ in serum red cell were no significant difference, but were higher (P < 0.05) than those of control group. The findings of this study showed that oral administration of Pu’er tea extracts could improve the growth performance and the capability of oxygen carry in passive smoking mice.

The resulting products were preliminarily purified and identified by LC-MS. As the result, the reaction yields performed without organic media was 4.1-5.6 times higher than that with organic medium (acetonitrile). In vitro antifungal assays were performed in Potato Dextrose Agar (PDA) culture medium, benzyl alcohol and phenylethanol exhibited antifungal activities against Colletotrichum Camelliae Massee obviously. Phenylethanol shown stronger antifungal activity with the EC50 of 0.71μL/mL and a minimum inhibitory concentration (MIC) of 1.8μL/mL. As for benzyl alcohol, the EC50 and MIC were 0.72μL/mL and 0.5μL/mL respectively. Benzylβ-glucoside also exhibited antifungal activities against Colletotrichum Cam elliae Massee obviously with concentrations from 5 mg /mL to 40 mg /mL in PDA culture medium.

Here we present a literature review on the relationship between different environmental parameters and d13C in plants. We will also consider the applicability of some of these relationships, as well as for the assessment of climate change dynamics and its implications.

The total DNA were extracted following a modification of CTAB method with some adjustments, influential parameters were optimized and the reaction system of PCR amplification was improved. On the basis of them, the genetic diversity of 13 poplar clones was studied by RAPD amplification with 18 primers. Each primer can produce 1 to 14 pieces of DNA fragments, of the total, 126 fragments were amplified. Among these, the number of polymorphic bands were 53, which occupied 42%, each primer can amplify 3 bands on average. It revealed that phylogenetic relationship was analyzed through cluster of bands in 126 amplified loci, RAPD analysis can examine lots of variations. Clone can be distinguished and purity can be measured by special bands. Molecular checking index was put forward according to special bands.

  Change of Trunk Sap Flow of Gingkgo（Ginkgo biloba L.）and its Response to Environmental Factors Liu Jian，Lai Nana，Zhao Bingxiang，Bian Xiuju 2007, 23 (6):  232-232.  Abstract + ( 1614 )   PDF ( 792228KB )   ( 614 ) In order to be master of the rule of water transpiration of gingkgo(G. biloba), and open out the mechanism of physiology action, The Thermal Dissipation Probe (TDP) was used in this experiment to monitor the dynamics of gingkgo (G. biloba) trunk sap flow in different growing periods, and an Automatic Weather Station was synchronously applied to measure the environmental parameters. The results indicated that change of sap flow was shown significantly different in fine day, cloudy day and in rainy day, daily cumulative flux of fine day>cloudy day’s>rainy day’s; The seasonal change trend of sap flow revealed that transpiration flux was the most in Jul., daily sap flow flux could reach around 23L.Daily sap flow flux fluctuating was relatively distinct in May, July and Sep., most distinct in Jul, discrepancy could reach 20L.According to average daily sap flow flux of different growth phases, transpiration flux of gingkgo of 12a was 2580L during 2006-growth season; Changes of sap flow flux in a growing season accorded with seasonal changes of temperature, soil relative humidity, soil temperature, relative humidity, PAR and rainfall. Stepwise regression analyses revealed that temperature was the main environmental factor influencing on sap flow, stepwise regression equation shows it reaches significant level.

The result was that karyotype characteristics of two hybrids 1894×Ps5 and XIN7×Ps5 and their parents were clarified, The karyotype were XIN7(2n=6x=42):2M+12sm+28m(2SAT)(2B)and 1894 (2n=6x=42): 2M+20sm+18m(2SAT)(2B)and Ps5(2n=4x=28): 2M+12sm(2SAT)+12m+2st(2B)and 1894×Ps5(2n=5x=35): 2M+19m+12sm+2st(B)and XIN7×Ps5(2n=5X=35):1M+21m+13sm(2B)； respectively. 【CONCLUSION】 the truth of hybrids were identified, the both were true, and the chromosome 1and the chromosome 1 of XIN7 parent and 1894 parent were in A-Genomen and B-Genomen, respectively; their ploidy was 6X. which Genomens chromosome 3 belonged to did not studied.

Our objectives were to examine the combining abilities and genetics parameters in 12 selected inbred lines in a diallel crosses to identify candidates for promising hybrid combinations and develop breeding strategy for germplasm improvement in early spring corn utilized in the northeast of China. The crosses were tested by random complete blocks methods, several agronomic traits in ear and kernel were evaluated, and data were analyzed in Griffing’s method IV for complete diallel crosses design. The combining ability analysis of diallel data showed highly significant effects for general combining ability (GCA) and specific combining ability (SCA) in tested traits of selected parents. Ear Diameter, Kernel Density and Kernel Rows of Ear have relative higher heritability and mainly controlled by additive effect; Grain Weight of Single Ear and Plot Yield have relative lower heritability, because non-additive effect take a larger portion in inheritance, these traits tend to affected by environment factors. Several parents can impact the corresponding trait in their offsprings remarkably, K12, Dan 340, HS169 can increase yield, P138 help to improve Kernel Density，local inbreds LK118 and Big Yellow Kernel conduce to prolong the length of ear respectively.

The results showed that content of microbial biomass carbon range from 200 to 600 mg/kg and decreased with increasing depth except for the conventional tillage and ridge, rice and rape. Microbial biomass carbon in no tillage and ridge, rice and rape in 0-10cm depth layer much more than other treatments after 14 years and showed significant differences；land use of rotation of waterlog rice and dry rape which was disadvantageous to increase the quantity of microbial biomass carbon value, which indicated microbial biomass carbon is good biological evaluation index of soil quality land use effect for paddy soil. Land use pattern of tillage and ridge, rice and rape is best for improve the Biological Activity and organic carbon of purple paddy soil.

The main results are as follow (1) GCA performance for 6 males was in the following order 042269>044130>05431-435>05421-425>042164>05416-420.The order for 13 female parents was De-3>De-2、De-16>De-1>De-6、De-23>De-18>De-7>De-25>De-30>De-8>De-20>De-10. (2)The best combination of SCA of yield was De-8×05421-425. And the fore three combinations of high yield were DE-16×044130，DE-16×042164 and DE-23×044130 in 78 combinations. (3) Genetic ability（h2B）of No. of pods per plant was highest .And lowest on weight -1000 seeds. De-3、De-2 and De-16 were good materials from field trial observe and statistic analysis.

The results showed: (1) There were the heterosis from mid-parent value, Hm, performing in most crosses, and the positive significant crosses were accounted for 25.00% of crosses for TN, On the contrary, there were 33.33%、25.00% and 8.33%of negative significant crosses for TN/Nic, TS and K2O respectively. There were 41.67%, 16.67%, 8.33% and 8.33% of positive significant crosses and 8.33%, 8.33%, 33.33% and 8.33% of negative significant crosses for Nic, Cl, TS/Nic and K2O/Cl respectively(2) There were 25.00%, 16.67% and 8.33% of crosses performing the significant heterosis over high-parent, Hh, for TN, Nic and Cl respectively, and there were no crosses with significant Hh for the others (3) There was no cross performing the significant heterosis less lower-parent, Hl, for the quality trails except for Nic, TS/Nic and TN/Nic with 8.33%、16.67% and 8.33% of significant crosses respectively.

A stub rape was divided into the bigger seed area, the middle seed area and the smaller seed area by seed weight; the relation of the grain weight of the different parts and seed vigor was significant. The seed vigor of three areas were reduced from big seed area to smaller seed area; 【Conclusion】The seeds of rape stem and some first effective branches were the bigger seed area which was the best area for rape seed selection and seed stock, and its seed vigor was the highest.

Large population was planted and the important family lines were selected by testing the yield in F3. Two new super-wheat lines, ShanMai107 with high yield and Shaanmai94 with strong-gluten, were bred. Shaanmai107 is 78cm, weak springness, multi spike, high percetenge of spike on the tiller, resistent to disease and lodging. Thousand kernal weight of ShanMai107 was 50g, and the yield reach 9712.5kg/hm2, which was 25.7% higher than the control cultivar Xiaoyan22. ShanMai94 is 75cm, weak-winterness, multi-spike, highly resistance to lodging, stripe rust, powdery mildew and leaf blight, and moderately resistant scab. one-thousand kernal weight of Shaanmai 94 was 51g and the yield was 8185kg/hm2, which was 6.0% higher than Xiaoyan22. The results showed that one of the effective way of breeding the ultra high-yield variety due to the materials of parent with abundant hereditary background, F2 expands the scale of planting, and examined yield in early generation.

The study on effects of sowing density, germinating conditions on the germination of male sterile Yunyan 87 (a flue-cured tobacco variety) pellet seed was conducted from 3 aspects ranging from sowing density, seedbed to watering. The results indicated that different sowing density posed greater effect on the germination of Yunyan 87’s pelleted seed than the germinating conditions did. When testing the seed germination, the suitable sowing density is 100 seeds per petri dish filled with 2 layers of filter paper soaked with tap water. Under these conditions, the germination force, the germination rate and the synchronization index of the pellet seed of Yunyan87 reached 76.0%，89.0% and 0.49 respectively.

By the agronomy measure, colony developments, cotton boll opening situation and the output constitution, the regression model of two objective properties was established, such as ginned cotton output and cotton ripe before frost. And the management index of cottons’ growing, and change relations between the output factors from the situation of mature bolls’ distribution and the influences were found. Then, different output levels of output composition were simulated. The main directions of attack on gaining yield output have been defined. The optimum cultivation measurements of shorter growing period cotton for high output high economic benefit were that: sowed in may 22-23, density is 7300-7500, N 11.04-12.0 kg, keeps the bearing-branch to count 8.4-8.7, combs the flower bud of lower part bearing-branch.

The result showed that , under the condition of experimentation , the relationship between the dry matter accumulation in aerial parts and hypogeal and the available accumulation temperature met the Logistic equation or met basically. The ranges of the effective accumulation temperature for the turning points of boom stage were: 981.6~1745.0℃ and 1655.4~2381.0℃ respectively. The highest instantaneous growth rate (Wmax) was 0.01862 g/（d ℃·plant）and 0.05976 g/（d ℃·plant） respectively ; and they appeared when the available accumulation temperature reached 1363.31 0C and 2018.22 0C respectively .

  Effect of Extrinsic ABA, Put and BR on Protective Enzyme Activities of Tomato Seedlings Under Sub-optimal temperature Bai Jie, Jiang Weijie, Yu Hongjun, Liu Yanpeng 2007, 23 (6):  317-317.  Abstract + ( 1881 )   PDF ( 207024KB )   ( 620 ) 【OBJECTIVE】Effects on protective enzyme activities of tomato seedlings under different concentration of abscisic acid (ABA), putrescine (Put) and epibrassinolide (BR) were investigated. 【METHOD】The tomato seedlings were pre-treated with various concentration of ABA、Put、BR, and then treated with sub-optimal temperature stress in the growth chamber. 【RESULTS】The results showed that under sub-optimal temperature the content of MDA increased; SOD activity raised firstly and then decreased. Pre-treated with ABA, Put or BR can decrease MDA content and improved protective enzyme activities. The appropriate concentration which deceased MDA content most is ABA1(0.2mmol/L), Put2(0.5mmol/L) and BR2(0.01mg/L).The most suitable concentration to SOD activities is ABA1(0.2mmol/L), Put3(1.0mmol/L) and BR2(0.01mg/L). Compared with the control the activity of SOD increased in 29.2%、17.4%、28.6% respectively and MDA content decreased in 47.7%、43.2%、42.2% respectively. 【CONCLUSION】The tomato seedlings were hurt by sub-optimal temperature. ABA、Put、BR played a positive role in alleviating the effect of sub-optimal temperature on tomato seedlings.

The lowest was treatment C, 7427kg. The difference between treatment C and D was obviously different, reaching to 5%. The highest content of sugar was treatment B, 15.5%; the lowest was treatment D, 15.2%.

Under phosphate deprivation conditions, the lipid composition of wheat (Triticum aestivum) shoot obviously changed by decreasing the relative proportion of PG. In addition, the increase of SQDG and DGDG were also observed. Although the decrease of PG was up to 49%, the photosynthesis characters and the amximal efficiency of PSⅡ photochemistry of shoot grown in phosphate-deficient conditions were similar to those of controls. These results suggested that the decrease of PG could be substituted for the increase of SQDG and accordingly kept normal photosynthesis. On the other hand, effects of phosphate deprivation on the composition of fatty acid of MGDG, DGDG, SQDG and PG were slight.

The results indicated that the main root development was statistically different in 96h or 144h tobacco seeding, however that was not was statistically different in 192h or 240h tobacco seeding. The number and total length of different genotype tobacco’s first, second, third and forth order lateral root was increasing with the increasing seedling age. The second lateral root, which total length was higher than any of the first, the third and the forth lateral root, was the main absorbing nutrient roots of tobacco seedling. The biomass from different tobacco seedling was different. The roots activity was different obviously and the chlorophyll of 5th leaf at the age of 50d seedling was not different statisticly. The cholorophyll content of Hongda was highest but that of NC82 was lowest of all tested variety.

There are five temperature which contains 19℃、22℃、25℃、28℃ and 31℃.When increase the temperature , the adult longevity and generation duration increased , the fecundity weakened . Under the five temperature , its adult longevity and generation duration were the longest at 19℃ and the shortest at 31℃. When feeding on the tobacco infected cucumber mosaic virus , its fecundity was the highest at 19℃ and the lowest at 31℃. Based on the parameters of the life table ,of the 5 temperature studied , the temperature at 22℃ is considered to be the most suitable for M. persicae (Sulzer).It was suggested that the development and reproduction of Myzus . (Sulzer) might be related to the temperature and tobacco infected cucumber mosaic virus.

The results displayed that the amount of dry matter and nutrients taken up by rice with NO3-N application were larger than that with NH4-N application in the early growing period of rice (20 days of growth). But with the increase of growth period, the application of NH4-N was demonstrated to be much more beneficial to the growth and nutrients uptake of rice. In addition, the rate of K applied was perceived to have influence on the effect of N fertilizer application. The growth and nutrients uptake of rice could be largely lessened by the application of K (K2O) with a rate of 0.51 g/kg, when the N rate was 0.15 g/kg. The sequence of N and K application also could have influence on rice growth and nutrients uptake, especially when high rate of K applied, which could result in the reduce of rice growth and nutrients uptake caused by the decrease of available N in soil.

According to the investigation during the seedling stage, the number of root were treated are more 1.8-2.9 than CK1, the plant height was not significant among all the treatments. The results showed that plasma treatment at the dosage of 1.0A×2 and 1.5A×2 could enhance nutrient uptake of soybean by chemical analyzed and yield surveying on dry matters during pod stage and maturity stage, the total fertilizer of N+P2O5+K2O was enhanced more than 10.0% compared to CK1,The use efficiency of N、P、K was enhanced by 25.2%~32.9%,3.0%~6.7% and 7.5~13.5%.It was clarified that plasma treatments at the dosage of 1.0A×2 could increase yield significantly by 11.0% than CK1.

Straw mulching could improve the contents of soil available nutrients【CONCLUSION】The content of hydrolytic N decreased smoothly after mature period. Although straw mulching was help to produce good quality Flue-cured tobacco,it should reinforce some effecttive cultivated adjustment research in order to make straw mulching become the best cultivation way to produce good quality Flue-cured tobacco.

By using SSR,20 apple cultivars were amplified by using 11 pairs of primers for genomic polymorphic analysis. SSR fragments of all pairs of primers were detected by polyacrylamide gel:163 loci were amplified and polymorphic percentage of amplified loci was 33.33%~63.16%. Based on the SSR results, calculation of the similarity coeffecient and also UPGMA cluster analysis showed that the apple cultivars could be classified into the four groups being similar to the traditional classification. And genotype map at a group of SSR loci of per cultivar was obtained. So genotype patterns of providing cultivars were firstly established. It would lay out a foundation for further genetype database, also indicating that SSR markers were suitable for early variety identification and plant breeder’s rights protection.

All of those physiological indexes would be considered as the system of heat-resistance select for the daylily.

Some bagges were bagged in different time(June10 and June23).Five types of bagges were used: Xiaolin bag(KM-2), Double layer paper inner black bag, Double layer paper inner red bag, Double layer inner plastic bag, Monolayer plastic bag. Researched bagged fruits′ respiration rate, total soluble sugars, titratable acids, firmness, et al. Respiration rate and water loss rate of all the fruits that bagged different bagges increased gradually to the peak in the first month of the storage. The respiration of the fruit that bagged MP is 47% heigher than the fruit bagged KM-2, at the same time soluble solids contents, titratable acids, firmness decreased gradually, respiration of all the fruits were strongly inhibited in the next month, fruit quality reduced slowly, then respiration of all the fruits increased gradually. The fruit that bagged MP bagges has higher respiration than others. The fruit that bagged in June23 has higher respiration than bagged in June10. In the end of the storage(120 days) the titratable acids, firmness of the fruit that bagged KM-2 bagges were 0.2087%、9.43kg/cm2, and the fruit that bagged Monolayer plastic bagges were 0.1812%、8.12kg/cm2. Results showed that the Monolayer plastic bag is worst. Xiaolin bag(KM-2) is the best after that is Double layer paper inner red bag and Double layer paper inner black bag, then is Double layer inner plastic bag .

Results suggested that nitrogen was the main cause of the nitrate content of pakchoi, between NO-3-N in nutrient and the nitrate content of pakchoi was positive correlative, but nitrate content would decrease with elevated NH+4-N concentrations in an appropriate concentration range. The nitrate content would decrease with elevated phosphorous and potassium at a certain concentrations. The relationship among nitrogen (NO-3-N, NH+4-N) phosphorous (P) and potassium (K) could also be an important factor. The nitrate content of pakchoi could be regulated and decreased by confecting nutrient.

The conclusion was that the vector has been transformed into EHA 105, which make a found of further study on delay-ripening of strawberry.

  Effect of Drought Stress on Physiological and Biochemical Chracteristics inThree Cultivars of Small Flowers in Chrydanthemum Qin Helan, Cao lei, Buyanhua 2007, 23 (6):  446-446.  Abstract + ( 1728 )   PDF ( 486513KB )   ( 541 ) Experiment was conducted with small flowers in Chrysanthemum of “A23” 、“A8”and“A27”.The changes of modality chraceristic of leaves, the relative water content (RWC) ,MDA content and dry weight were studied under drought stress.The results indicated that the wilted time , the decreased extent and process of the relative water content (RWC) were all different in three cultivars.The content of MDA raised first,declinedd then,and increased continually later.The growth was restrained both in leaves and roots,but the value of root/crown was raised under drought stressed. The conten of dry material was different in the organ and cultivars.Integrated analysis showed that A27 had the strongest adatability under drought stress with the late time of wilted ,slower decreased process of RWC, low extent and content of MDA,and little range of dry material content changed.

Calcium fertilization can enhance the calcium quantity in vegetative food, thus it would do favor to the health of the human being. If the new root growth is blocked or the transpiration reduces, and the calcium content in the crops suddenly reduces, it will affect the regular growing development of the crops. Through spurting Calcium fertilizer on the out root of the crops, it can have a fast effective supplement of the calcium need to the plant. This experiment tests the following conclusion through three foliage sprays continuously with different calcium content during blossom period, fruiting period and inflated period: comparing the apple output per hectare with spurting clear water, if spurting Calcium fertilizer with 160g/L, it can increase production 5666.6kg/hec, the rate is 14.4% and the analysis of variance is shown extremely significant level. If with 120g/L, it can increase 4888.8kg/hec, the rate is 12.4%, and the analysis of variance is shown significant level.

  Influences of plant growth-promoting rhizobacteria CH1 on phenolic metabolism in cucumber and its relation to resistance to cucumber damping-off Liang Jiangen,, Zhang Bingxin, Yu Jingquan 2007, 23 (6):  462-462.  Abstract + ( 1364 )   PDF ( 270466KB )   ( 776 ) In order to explore the mechanism of the resistance to cucumber damping-off induced by plant growth-promoting rhizobacteria CH1, the physiological and biochemical method was employed to study the changes in activities of Phenylalanine ammonia-lyase(PAL), Polyphenol oxidase(PPO) and contents of ferulic acid, chlorogenic acid, lignin, phenolic compound and flavonoid of phenolic metabolism in cucumber roots pretreated with plant growth-promoting rhizobacteria CH1 and infected by Pythium aphanidermatum. The results showed that the activities of PAL and PPO, the contents of ferulic acid, chlorogenic acid, lignin, phenolic compound and flavonoid in roots treated by CH1 increased higher than those of the nontreated when plants were infected by Pythium aphanidermatum. This indicated that CH1 could induce resistance of cucumber to Pythium aphanidermatum by increasing its enzymes activities of phenolic metabolism.

A number of quorum sensing systems have been found in Gram-positive and in Gram-negative bacteria, which control the expression of multiple genes. This review focuses on the latest research and function of quorum sensing, and describes the prosperity of application in biotechnology.

Different effects of protoplast formation of Men-myco-93-63 were tested, including the sub-culture medium, the sub-culture time, the concentration of Lysozyme or Gly, et al. Methods for the formation and regeneration of Stremptomyces roseoflavus Men-myco-93-63 protoplasts are described: Spores shake cultured in TSB for 24h,Add ω＝2%Gly to SGGP(culture medium), continue shake cultured for 40h, Collect mycelium, Protoplasts of Stremptomyces roseoflavus Men-myco-93-63 strains were obtained by treatment with lysozyme whose concentration is 2mg/ml in a protoplast formation buffer. The protoplast of Men-myco-93-63 has been prepared successfully and Cells were regenerated on a complex medium. regenerated with 4.8×106/ml on R5.The methods were applicable to other species of Stremptomyces roseoflavus as well. It laid a foundation for protoplast transform and gene expression.

Based on the studies and investigation in paddy fields from 2000 to 2005, the pattern of occurrence, key factors affected its population dynamics and rice yield losses were preliminarily proved, and its control threshold was recommended at peaks of adults occurred and eggs hatched, when 10 eggs were found in 100 seedlings or more than 1% plants were damaged at seedling stages and average 1 egg laid in one hill or 3-5% plants were infested. Field investigation on distribution pattern of rice stem maggot showed that the eggs distributed evenly and the larva aggregated, and then the sequential sampling and simplified field investigation techniques, and population monitoring and forecasting methodologies GIS-based were proposed and the systems of data pool management, population stepwise regression, and the simulating model of population dynamics were constructed. By improving the population monitoring and forecasting methodologies, the modified systematical management strategies, focused on the first generation to suppress the population number of second generation, integrated with agricultural, physical and chemical managements, had played significant role in decreasing the population density of rice stem maggot and increasing the economic, social and ecological profits in southern China.

The Control effect experimentation in land was used 1500 times liquor. The Control effect contrast experimentation of small parts of land on Empoasca flasvescems Feb. were used 500,1000,1500 times 0.2 percent celangulin. 【RESULTS】 The result of the experimentations indicate that the part used 500 times liquor’s death rate was 98.67% and the part used 1000 times liquor’s death rate was 85.33% and the part used 1500 times liquor’s death rate was 73.33% indoor after 3 days. And in land, using 1500 times liquors 10 days on Ectropis obipua hypulina Wehrli, the control effect is 88.13% to 90.69%. Using 500 times liquor’s 8 days on Empoasca flasvescems Feb., the control effect is 63.66% to 80.11%. 【CONCLUSION】 The experimentations indicate that the control effect of the liquor is good and the control effect lasts long. 0.2 percent celangulin, the botanical pesticide, can be used in a nuisance -free tea base.

  Changes of Peroxidase and Isoenzyme in Tobacco Infected by Pseudomonas Solanacearum Hu Qingping,, Xu Jianguo, Bo Fangfang, Chen Wuling 2007, 23 (6):  497-497.  Abstract + ( 1496 )   PDF ( 260029KB )   ( 833 ) 【OBJECTIVE】 Comparatively peroxidase (POD) and isoenzyme of roots, stems and leaves in tobacco infected with Pseudomonas solanacearum were studied in order to offer academic foundations for breeding resistive plants. 【METHOD】 Colorimetry for POD activity and polypropylene amic gel electrophoresis (PAGE) for isoenzyme were used. 【RESULTS】 After inoculated by Pseudomonas Solanacearum, it was found that there was higher peroxidase (POD) activity in leaves than that in roots and in stems, and the POD activity in roots, stems and leaves was highest in 3 days but in leaves which improved 29.67% percent than those form the contrast. Isoenzyme electrophoresis showed that difference occurred in numbers and types of isoenzyme bands and related activity of every band between different samples. 【CONCLUSION】 POD was a protectal enzyme and it was changing with infection, so it was suggested that POD may have close relation to resistance against disease.

The ethanol extracts of Zanthoxylum bungeanum、Corydalis bungeana、Oldenlandia diffusa et al.were obtained with ultrasonic wave from 40 Chinese material medicines, the insecticidal activity of these extracts to Myzus persicae was tested in laboratory. Results showed that the 12 species of them presented more than 50% insecticidal activity to Myzus persicae, the extracts of Zanthoxylum bungeanum and Corydalis bungeana had the best insecticidal activities, the corrected mortality could reach 93% and 83% respectivly after 48 hours. Zanthoxylum bungeanum and Corydalis bungeana et al. 12 Chinese Material Medicines were worth developing.

By studying the topic ,It will be clear about the jasmine harmful insect's type, to grasp distribution and harms and rule of the main harmful insect in the Yuanjiang planter area. From this , economical, effective, and safe preventing and controlling technology strategy has been proposed ,which will solve the jasmine production difficult problem well.Experiment has indicated that, Dealing with the main pest of jasmine—jasmine flower bud snout moth , only suitably selects highly effective, low poisonously, low residual agricultural chemicals. After experimental sieving, The biological agricultural chemicals, such as Tiantai of 2.1%1000 times liquid, Aifuding of 24.5%1500 times liquid, Abading of 1.8% 3000 times liquid, Avermectin of 1.8% 3000 times liquid, Jinfuding of 1% 1000 times liquid and so on.,are at present ideal medicament for preventing and controlling jasmine flower bud snout moth .These medicaments have displayed the better preventing and curing benefit in the big area production utilization.The best spraying time is suggested from 8 to 9 o’clock at night, which may remarkably enhance the preventing and curing effect.

This article uses the Hefei data from 2000 to 2004, calculated the ecological footprint(EF), ecological capacity(EC),ecological deficit(ED) and Ecological Footprint per ten thousand Yuan GDP. The result indicated the EF assumes the trend of escalation and greatly higher than the EC in Hefei, the phenomenon of ED is serious. In 2004,ED per person was 2.10108hm2;when in 2000,it was just 1.7041hm2. The balance between supply and demand relies on other area input. But ten thousand Yuan GDP Ecological Footprint assumes the drop tendency. In 2004 ten thousand Yuan GDP ecological footprint was 1.797977,when in 2000,it was just 2.545955. It indicated that resources using and the efficiency has enhanced with the industry and economical growth process.

  Potentiality Evaluation of Regional Cultivated Land Consolidation and Rehabilitation Based on Entropy Weight method: A Case Study of Chongqing Municipality Li Ping, Wei Chaofu, Qiu Daochi 2007, 23 (6):  536-536.  Abstract + ( 1372 )   PDF ( 621423KB )   ( 758 ) Based on constructing evaluation index system for regional potentiality of land consolidation and rehabilitation, potentiality of cultivated land consolidation and rehabilitation in Chongqing municipality was quantificationally evaluated using entropy weight method and synthetical evaluation method. The results indicated that indicator weight was followed such a sequence: input-output rate>increasing ratio of forestland proportion >increasing ratio of cultivated land output> increased cultivated land coefficient. The potentiality of cultivated land consolidation and rehabilitation was divided into four grades, and it was highest in ecological economic zones in Three Gorges reservoir area and higher in Western Chongqing Region. Western Chongqing Region, whose development rate and input-output rate were higher, played an important role in the economic development of Chongqing, thus the advantages of cultivated land consolidation potentiality were prominent. The ecological benefits of Three Gorges reservoir area were so high that cultivated land consolidation and rehabilitation could achieve the sustainable development.

But, with the development of industry and agriculture, soil pollution becomes more serious, and soil animals are influenced. So, this paper summarized the ecological influences of four environmental stresses on soil animals as follows: (1) pesticide pollution: with the increase of pesticide pollution, community of soil animals, the total number of the individuals, the diversity index, the homogeneous index, the density of individuals were decreased. But the influencing extents of different types of pesticides on soil animals were different, the insecticide was more serious than herbicide. Different soil animals were sensitive to different types of pesticide. (2) heavy metal pollution: the influences of heavy metal pollution on soil animals were similar as pesticide pollution. (3) N deposition: the influences of N deposition on soil animals changed with the specific environment. (4) atmospheric CO2 enrichment: some foreign researchers mainly used the FACE(Free-Air CO2 Enrichment)system to study soil animals, they found that soil animals may generate positive, neutral, negative response. Finally this paper pointed out several new areas which should be further studied in soil animal ecology in China.

The results summarized as follows: that most secondary structure was found in rosemary’s root structure, which was madeup periderm and vascular tissue , and there are many big vascular vessel. The periderm in stem is so loosen and fewness stomas are covered. The upgrowth xylem and thicky medullary rayare also found in the stem. The dense epidermal hair and round less opening stomas are spreaded on lower epidermis. It also found deepth stratum corneum on the leaf surface. In internal structure, there are growthy vascular bundle and palisade tissue etc. The anatomy structure of rosemary adapted to the ecological environment, so it embodied the dialectica lrelation between structure and function.

The results showed: the rice would be damaged by low chlorimuron-ethyl concentration. When the amount of chlorimuron-ethyl≤0.5mg/kg, the bio-fertilizer could degrade it significantly. It could impair the polluted soil, promote the growth of root, increase the grains per panicle of rice, panicle number of per hill and increase the yield. When the chlorimuron-ethyl amount was 6mg/kg, bio-fertilizer could not degrade the herbicide. From the comparison of every treatment, the best economy amount of bio-fertilizer was 30kg/hm2.

We anaysised the difference between the treatment and CK. The result shows that there is no significant difference between the treatment and CK, and it is feasible to plant winter wheat with reclaimed water irrigation.

The data shows that households handle crop straw through ways of burning and utilizing. Through analysis of Logit modle, we find that the peasant households is affected by characteristics of heads of households, characteristics of families, characteristics of producing and region in choosing handling ways of crops straw.

The results indicated that two kinds fertilizer all improved the soil available Pb and Pb accumulation in root of wheat. The low dosage of N fertilizer （0.1~0.3g／kg）promoted biomass of wheat and the higher dosage of applying fertilizers （0.5~0.7g／kg）prohibited the growth of the wheat .Applying N fertilizers decreased the Pb accumulation in up part of wheat. While there was no obvious correlation between soil available Pb and Pb contend adsorbed by wheat . The N fertilizer can effectively alleviate negative influence of Pb on wheat seedlings and can inhibit the activity of POD enzyme in wheat. Between urea and NH4HCO3, the low dosage of urea fertilizer was better than NH4HCO3.

Based on the research we integrated, we evaluated the ecosystem services in Wuhu, depending on summarized parameters. It was found that the value of ecosystem service decreased 74 million Yuan from 1996 to 2004 with GDP increasing 22707 million Yuan. Therefore, we suggest that ecological construction should be taken into account in the land use frame of Wuhu.

Environmental problem has become one of the main obstacles to sustainable tourism development. The tourism environmental impact in the Lihu parks was researched. Influences of tourism activities and recreational building on environment, and influences of environment on tourists were included in the paper. The results were as follows: (1) The relationship was Leq =19.91+7.08191nN between the noise of tourism (Leq, dB) and the number of tourists (N). (2) The vegetations of Lihu parks could purify atmosphere efficiently. The rates of removal NOX, SO2 were 7.7%~11.7%, average 32.3%, 28.1%~37.5%, average9.9%, respectively. (3) The most of tourists were satisfied with sound environment.

The experimentation indicated that Cymbidium sinense pollen original in Hainan Province readily germinated in artificial liquid mediums in the experiments with long pollen tubes. The average germination percentages of seven treatments up to 78% except the contrast treatment. It was 83.99% on liquid medium containing 10% sucrose, 20mg/L boric acid and 5 mg/L calcium chloride with optimizing result. For simple principle, it will be used for identifying Hainan Cymbidium sinense pollen viability in future.

The 200 polymorphic bands of RAPD bands were obtained.The average of polymorphic bands of per primer was 11.8. The RAPD based genetic similarity ranged from 0.356 to 0.676. UPGMA cluster analysis based on RAPD molecular markers data divided 10 species into four groups. The results showed that the RAPD approach was more efficient on Study of genetic background and relationship of Dendrobium germplasms .

In this paper, resistance genes in maize genomic database and references were explored, and the consensus map was constructed with the aid of high-density linkage map IBM 2005 neighbors and the adjacent markers to each major gene or QTL. Consequently, a total of 78 major genes or QTLs involving in resistance to 14 maize diseases were screened out. The results demonstrated that the number of resistance gene distributed on 10 chromosomes asymmetrically, large number of major resistance genes was located on chromosome 3, 6 and 10, and few on chromosome 5 and 7, and resistance gene cluster were found obviously. The results would be valuable for identification of new resistance gene and construction molecular marker-assisted selection strategy for disease resistance in maize.

  Antioxidative System Characteristics of Sweet Potato Transferred both Cu/Zn Superoxide Dismutase and Ascorbate Peroxidase Gene under Water Stress and Rewatering Li JianMei, Deng XiPing 2007, 23 (6):  616-616.  Abstract + ( 1427 )   PDF ( 357546KB )   ( 546 ) Two strains, Cu/Zn SOD and APX gene transferred sweet potato (TS) and non-transgenic sweet potato（Ipomoea batatas L.）（NT），were used as experimental materials to study the effects of Water stress and rewatering on antioxidative system characteristics of sweet potatoes. The result showed that the activities of SOD of both TS and NT first increased and then decreased under water stress, but the activity of SOD of TS was always higher than that of NT, under 24h water stress, the activity of SOD of TS was 20% higher than that of NT. The activities of SOD of TS and NT both decreased after rewatering. Under water stress, the activity of APX of TS firstly increased and then decreased; in NT it firstly decreased and then increased. After rewatering, the activity of APX of both TS and NT firstly increased and then decreased, after12h rewatering, the activity of APX of TS was 50% higher than it of NT (Fig.1). When PEG treatments were imposed for 24h and 36h in TS, the AsA content increased by 12% and 53%, however, the AsA content was in NT increased firstly and then decreased. After PEG treatments for 0h, 12h and 24h, AsA content in TS was higher by 3 %, 18 % and 37 %, than that in NT (p<0.05), respectively (figure 2). Recovery by no PEG solution, AsA content decreased in both TS and NT. Recover for 12h, 24h, AsA content in TS was 45% and 37% higher than that in NT (p<0.05). Under the no stress and PEG treatments for 12h and 24h, CAR content in NT was higher by15 %,21% and 13 %, than that in TS (p<0.05), respectively. After PEG treatments for 36h, CAR content in TS and NT was almost the same. Recovery in on PEG solution, CAR contents decreased in TS and NT. Recovered for 12h, CAR content in NT was 11% higher than in TS Under water stress, the membrane structural stability of TS was better than it of NT, after 24h water stress and 12h rewatering, the MDA content of NT were 20% more than it of TS (Fig.5). The result showed that Transferred both Cu/Zn SOD and APX gene，the antioxidative system of the transgenic sweet potato had a higher ability to protect and recover the antioxidative system of the plant.

Carps were randomly divided into treatments which contained 3 replicates and 30 carps for each replicate. The basal diet which was added 0 mg taurine per kilogram was used as control group, and the experiment lasted for 56 days. The results showed that growth performance of carps were remarkably increased than control group. At 800mg/ kg level , biology synthesis evaluate(BSE) got the maximum and feed coefficient (FC) got minimum ( P< 0.05) ; In the adding level of taurine at 800～1200mg/ kg , activities of protease, lipase and amylase in hepatopancreas and intestine of carps were remarkably enhanced ( P< 0.05) . According to above results, the optimum dietary taurine level to caps was 800mg/ kg.

  Comparative Studies on Morphological Differences among Four Local Populations of Mitten Crab Wang Wu, Xu Can, Zhang Wenbo, Ma Xuzhou 2007, 23 (6):  648-648.  Abstract + ( 1532 )   PDF ( 589674KB )   ( 643 ) Morphological differences among two Eriocheir japonicus populations collected from Suifen River and Hepu River and two Eriocheir sinensis populations from Changjiang and Liaohe River were studied based on cluster analyzing and discriminal analyzing on 17 morphological characters , and the hybrids of Eriocheir sinensis (♂) × Eriocheir japonicus(♀) was also researched as an assistant subject. The result of cluster analyzing showed that the five populations can be divided into two groups:one group concluded Suifen,Hepu, Liaohe and the hybrids ,and the Changjiang population was single. The discriminal analyzing also separated samples from changjiang and the other four populations. The mean accurancy rate of discrimination of female crabs and male crab were 74.2% and 75.4％ . In the long period of process of evolution, the mitten crabs in China mainland gradually came into being different local populations which greatly rely on the geological environment around.These local populations possessed strongly regionality on the growth. Eriocheir japonicus and Eriocheir sinensis could hybridize and breed naturally, so reproductive isolation principles didn’t exist between them.

Taking Guangdong Province as an example, the ratio of the gross output value of marine industries between the province and the country was 17.7 percent in 2005, the main industries, the superior industries and the industries should be developed of which were the coastal tourism, the marine electric power and seawater utilization, the offshore oil and natural gas and so on. Guangdong Province will become a powerful province of marine economies, the ratio between the added value of marine industries and the GDP of which will be 15.0 percent in 2010.

Under such circumstance, studies of the possible space for agriculture in Beijing and how to use it reasonably are necessary. 【METHOD】Based on the analysis of influencing factors of agriculture development, Newton's theory of space as a guide, and Venn diagram as the display means, Beijing's agricultural development space was explored. 【RESULTS】Beijing's agricultural development space can be divided into two kinds of spaces and four subspaces, the former consists of the absolute space and the relative space, and the later comprises the reality space, the extensible market space, the extensible policy space and the cooperative development space. For different agricultural spaces, the content, feature and use directions are different accordingly. Measures should be carried out to stabilize the absolute space and to explore the relative space, at the same time, we also need to use the reality space efficiently, exploit the extensible market space, the extensible policy space, etc, to break through the restriction of agricultural resources in Beijing. 【Conclusion】The study can be beneficial to the protection and expanse of agricultural development space, and to promote 调和sustainable agriculture development of Beijing.
