Optimization of Phenol-Sulfuric Acid Determination Conditions of Polysaccharide in Polyporus Umbellatus by Orthogonal Test

Abstract: 【Objective】To establish rapid quantitative analysis method for polysaccharide in Polyporus umbellatus.【Method】Taking phenol - sulfuric acid method to content of glucose as parameters. The absorbance is measured at 490 nm. L9 (34) Orthogonal Design optimum determine conditions. 【Results】when the sample solution used 1.0ml, 0.8 ml of 5% solution of phenol, and sulfuric acid 5.0ml, 100 ° C water bath for 15 min reaction conditions determination, the results is best. Also sulfuric acid increment to result influence biggest, the water bath temperature is next, the phenol quantity influence is smallest. The glucose had good linear relationship in 5.0~40.0μg/ml. RSD=1.58% precision (n = 6), measured 3h good stability. The average of recovery was 99.22%, RSD=1.87% (n = 3). The content of polysaccharide in Polyporus umbellatus was measured 3.46%, RSD=2.07% (n = 4). 【Conclusion】This optimization method is highly sensitive, simple, stable data, accurate, good reproducibility and worth promoting.
