Establishment of indirect ELISA method to detect Porcine Circovirus 2 antibody

Abstract:

The research established a fast and sensitive indirect ELISA method to detect PCV2 antibody. The experiment used the prokaryotic expressive PCV2-Cap protein as the envelope antigen. In addition, we also confirmed the optimal coating-antigen concentration、Rabbit Anti-Pig IgG concentration, and selected the best detection-pending serum dilution、critical value, and detected specificness、stability、coincidence rate by indirect ELISA method. The results indicated only the PCV2 postive serum was detected by indirect ELISA; The variance among the three repeat experiments was nonsignificant; The coincidence rate between indirect and the PCV2 Kit of JBT in Korea was up to 92.5%; The 204 swine serum samples from the Fujian Province were detected by indirect ELISA, the positive rate was 76.5%. The experiment revealed the optimization indirect ELISA was of good specificity, good repeatability and stability, and may be used as the serodiagnosis and the epidemiological study of PCV2 antibody.
