水曲柳愈伤组织诱导及悬浮细胞体系的建立

Callus induction and establishment of cell suspension culture system for Fraxinusm andshu rica

摘要: 水曲柳悬浮细胞培养比较困难,本研究首次获得水曲柳悬浮细胞。以水曲柳叶柄为材料诱导愈伤组织,通过2,4-D浓度的筛选,建立稳定的水曲柳悬浮细胞培养体系,可为水曲柳悬浮细胞系遗传转化或诱变后单克隆的筛选提供材料。水曲柳愈伤组织在WPM+BA0.1mg/L+2,4,-D0.015mg/L+20g/L蔗糖液体培养基中振荡培养3-4代,可获得稳定的悬浮细胞系,最佳继带周期为15d。

Abstract: A stable suspending cell culture of Fraxinusm andshu rica could be used for genetic transformation or screening monoclone after mutagenesis. In this study, stalks of Fraxinusm andshu rica were used to induce calli and suspending cells were obtained after a series gradient of 2,4-D were added in the culture. The calli could be subcultured 3-4 gengerations every 15d in the media of WPM+BA0.1mg/L+2,4,-D0.015mg/L+20g/L sucrose to get stalbe cell culture.
