猪嵴病毒福建株结构蛋白VP 基因的克隆与生物信息学分析

Cloning and Bioinformatics Analysis of the VP Gene of Porcine Kobuvirus

摘要: 为丰富猪嵴病毒结构的分子流行病学数据,明确福建株猪嵴病毒结构蛋白VP的特征,研究运用RT-PCR方法设计特异性引物,对猪嵴病毒结构蛋白进行分段扩增后进行克隆测序,并将测序结果进行拼接后进行生物信息学分析。结果表明,所扩增福建株猪嵴病毒含有完整的结构蛋白VP,其基因全长为2529 bp,编码有843 个氨基酸,理论等电点为5.34,理论分子质量为89.603 kD,不稳定系数为37.71,最大疏水指数为2.522,最小疏水指数为-2.622。其和XX株猪嵴病毒结构蛋白核苷酸同源性最高,仅为86.7%,与swine/S-1-HUN/2007/Hungary 株猪嵴病毒结构蛋白核苷酸同源性最低,为83.7%。从遗传进化上看,猪嵴病毒结构蛋白在遗传进化上呈现2 个独立的分支,中国猪嵴病毒分离株在2 个遗传分支上均有分布。

Abstract: To enrich the gene for the newly identified porcine kubovirus, theVP gene of porcine kobuvirus was amplified by RT-PCR method from tissues of piglets associated with diarrhea in Fujian Province and then sequenced, then bioinformatics analysis were conducted to investigate the structure and function of the porcine kubovirus VP gene. The results demonstrated that the porcine kobuvirus VP gene was 2529 bp in length, coding an open reading frame (ORF) with 843 amino acids. The isoelectric point, molecular weight and instability index of porcine kubovirusVP protein were 5.34, 89.603kD, 37.71, respectively. The maximum and the minimum hydrophobicity were 2.522 and - 2.622, respectively. Compared with the PKV VP genes published previously download from GenBank, the sequenced gene shared the highest homogeneity with XX strain, with 86.7%; while had the lowest homogeneity with swine/S-1-HUN/2007/Hungary strain, which was 83.7% . The porcine kubovirus VP gene shares two different phylogenetic relationship branches, and the Chinese porcine kubovirus isolates distribute at both branches.
