新疆野扁桃自交不亲和基因SSK1 的克隆及生物信息学分析

Cloning and Bioinformatics Analysis of Self-incompatibility Gene SSK1 of Wild Almond in Xinjiang

摘要: 为了获得新疆野扁桃居群中自交不亲和花粉特异性表达的SSK1 基因全长,以新疆野扁桃植物叶片及花粉为试验材料通过rt-PCR 技术以及生物测序等技术,克隆得到了一个新的SSK1 基因全长cDNA,PetSSK1。PetSSK1 全长为602 bp,开放阅读框为534 bp,共编码了177 个氨基酸,经生物信息学分析,PetSSK1 蛋白的分子式为C910H1421N233O295S6,相对分子质量为20538.0,理论推导半衰期为30 h,等电点为4.52,不稳定指数为41.48,平均疏水指数为-0.567,PetSSK1 蛋白属于水溶性不稳定蛋白。首次从新疆野扁桃植株中克隆得到了SSK1 基因全长。通过对该基因的克隆及序列分析,为今后的表达分析以及有效利用该基因获得自交亲和植株奠定了基础。

Abstract: The research aims to obtain the full- length SSK1 gene with specific expressions in the selfincompatible pollens from wild almond clusters of Xinjiang area. In this research, the leaves and pollen of wild almonds of Xinjiang area were chosen as the materials. Through the rt-PCR and bioinformatics sequencing, the author got a new full-length SSK1 gene, named as PetSSK1. The results showed that the full-length of PetSSK1 was 602 bp and the open reading frame was 534 bp, encoding 177 amino acids. Through the bioinformatics analysis, the molecular formula of PetSSK1 protein was C910H1421N233O295S6 and the relative molecular mass of it was 20538.0, and its estimated half-life was 30 h in theory. Moreover, the result showed that the hydrophobic index and the grand average of hydropathicity of PetSSK1 protein were 41.48 and - 0.567, respectively, indicating that PetSSK1 protein belonged to the unstable water-soluble protein. It is the first report that a novel SSK1 gene has been cloned from wild almonds of Xinjiang area. Through the gene cloning and sequence analysis, expression analysis laid the foundation for the future and the effective use of the gene to obtain selfcompatible plants.
