融合蛋白ABF 对金葡菌感染版纳微型猪近交系皮肤创伤的修复作用

Protection Effect of Fusion Protein ABF Against Staphylococcus aureus in Banna Mini-pig Inbred Line Wound Infections

摘要: 随着金葡菌对畜牧业破坏性增加和耐药性增强,人们对研发靶向金葡菌的有效保护性疫苗日趋迫切。为开发出更具免疫保护效力的抗菌疫苗,本研究首次利用EsxA、EsxB联合细菌鞭毛蛋白Flic,即制备出融合蛋白ABF,并在版纳微型猪近交系皮肤创伤模型上做实验验证。实验用版纳微型猪分为4组:对照组、模型组、实验组Ⅰ和实验组Ⅱ。结果发现单独使用EsxB蛋白与对照组微型猪的免疫指标之间差距并不显著,而预先经融合蛋白ABF免疫的微型猪,检测到其外周血免疫因子IgG 总量表达显著上升,即机体体液免疫水平提高,同时融合蛋白ABF 可以激活T 细胞。此外其脾脏淋巴细胞CD8a、CD3e 和IFN-γ阳性率均显著增高,皮肤创伤修复速度加快。总之,本实验研究成果证实融合蛋白ABF 可作为潜在有效的金葡菌亚单位疫苗,为防治金葡菌提供新的研究途径。

Abstract: Staphylococcus aureus (SA), one of the major causes of livestock infection, is resistant to many kinds of antimicrobials. It is urgent to develop vaccines targeting at this pathogen. In this study, a fusion protein (ABF) composed of EsxA, EsxB and Flic was firstly generated. To examine the effect of ABF against SA, Banna mini-pig inbred line wound models were established and divided into four groups, the control wound group, the SA infected group, the EsxB pre-immunized group and the ABF pre-munized group. The results showed that peripheral IgG expression level in the mini-pig pre-immunized with ABF was significantly higher than that of the other three groups. Besides, ABF could activate T cell as well. The ratios of CD8a, CD3e and IFN-γ positive cells to spleen lymphocytes were all increased in the ABF group. In conclusion, all the results showed that ABF could significantly induce chemokine response and was functional during wound healing.
