渗透胁迫对蒙古沙冬青试管苗生理特性的影响

Effect of Osmotic Stress on Tube Culture Physiological Characteristic of Ammopiptanthus mongolicus ( Maxi m) Cheng.f

摘要: 通过调节MS培养基中蔗糖浓度形成的不同渗透势,模拟干旱胁迫条件,对不同渗透胁迫条件下蒙古沙冬青组培苗丙二醛(MDA)、可溶性糖含量、过氧化物酶(POD)活力和脯氨酸含量的变化情况进行试验研究。结果显示,随着培养基蔗糖浓度的增加,渗透胁迫程度也不断加强,丙二醛和可溶性糖含量明显增加,过氧化物酶活性和脯氨酸含量呈现先上升后下降趋势。15%蔗糖浓度的培养基其渗透势为-1.540MPa且胁迫72h,是沙冬青组培苗的耐受阈值,其脯胺酸含量及过氧化物酶(POD)活性均明显下降,幼苗生长停滞。

Abstract: This paper simulated drought stress conditions by adjusting the sucrose concentration and forming different osmotic potential in the MS medium, and through the testing, studied change rule of malondialdehyde (MDA), soluble sugar content, peroxidase (POD) activity and proline content of Ammopiptanthus mongolicus ( Maxi m) Cheng.f’s tube culture under the condition. The results show that with the increase of drought stress, the content of MDA and soluble sugar increases significantly, and activities POD, proline content roses at the beginging and then declines. Therefore, ψπ=-1.540Mpa and stressing 72h is the tolerate threshold of stress,and it causes the obvious decline of proline content and POD activities, and seedling growth is affected seriously.
