陇东关山区域独活质量评价及成分测定

Quality Evaluation and Component Determination of Angelicae Pubescentis Radix in Guanshan Region of Eastern Gansu

摘要:

本研究旨在对陇东关山区域中药材独活开展质量评价与成分测定,以期为今后独活种质资源研究利用提供参考。运用指纹图谱相似度评价软件建立陇东关山区域13批独活HPLC指纹图谱,采用系统聚类分析(HCA)、主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)对13批独活进行质量评价;同时测定独活主根和侧根中二氢欧山芹素、当归醇B、当归醇A、佛手柑内酯、二氢欧山芹醇醋酸酯、蛇床子素、二氢欧山芹醇当归酸酯8个成分的含量。13批独活共标定了18个共有峰,指认了8个成分,相似度为0.911~0.997,利用HCA将13批独活样品分为4类,PCA和HCA分析结果基本一致,OPLS-DA筛选出5个起主导作用的成分;指认的8个共有峰在独活主根和侧根中含量不同,侧根中蛇床子素、当归醇A的含量明显高于主根,而主根中二氢欧山芹素含量高于侧根,异欧前胡素则无显著差异。建立的HPLC指纹图谱、化学计量分析及多成分测定方法稳定可靠,能为本区域道地药材质量控制提供理论指导。

Abstract:

Quality evaluation and component determination of the Chinese herbal medicine Angelicae Pubescentis Radix (Duhuo) in Guanshan Region of Eastern Gansu were conducted to provide references for future research and utilization of Angelicae Pubescentis Radix germplasm resources. HPLC fingerprinting of 13 batches of Angelicae Pubescentis Radix from this region was established using similarity evaluation software, and quality assessment was performed through hierarchical cluster analysis (HCA), principal component analysis (PCA), and orthogonal partial least squares-discriminant analysis (OPLS-DA). The contents of eight components in both primary and lateral roots of Angelicae Pubescentis Radix were simultaneously determined, including columbianetin, angelol B, angelol A, bergapten, columbianetin acetate, osthole, and columbianadin. The 13 batches showed 18 common peaks (with 8 identified components) and similarity indices ranging from 0.911 to 0.997. HCA classified the samples into four categories, and the result was consistent with PCA findings. OPLS-DA screened five dominant components. The eight identified components exhibited differential distribution between primary and lateral roots: osthole and angelol A were significantly higher in lateral roots, whereas columbianetin was more abundant in primary roots. There was no significant difference for isoimperatorin. The developed analytical method integrating HPLC fingerprint profiling with chemometrics and multi-constituent determination proved to be robust and reproducible, offering scientific support for quality standardization of locally sourced Angelicae Pubescentis Radix.
