‘夏日阳光’番茄侧芽离体培养快繁技术

In vitro Culture Technique for Rapid Propagation of Lateral Bud of Lycopersicon esculentum‘Xiariyangguang’

摘要: ‘夏日阳光’番茄是近年来引进的高端品种,丰产性好、品质高,深受市场的青睐,但其种子为F1代杂种,价格昂贵,很难在市场上大面积推广。利用植物离体培养技术快速繁殖‘夏日阳光’番茄种苗,是生产上推广该品种的一条可行途径。为快速建立‘夏日阳光’番茄无菌株系以及获得其试管芽苗的高效增殖,以‘夏日阳光’番茄侧芽为材料,探讨其离体培养过程中的关键技术。试验结果表明,采用0.1% HgCl2溶液(w/v)加2滴土温20消毒8 min的处理方式消毒后,培养在MS 0.1 mg/L BA 0.1 mg/L IBA培养基中,可以较快速地建立‘夏日阳光’番茄无菌株系,成苗率为46.7%;继代增殖适宜的培养基配方为MS 0.5 mg/L BA 0.1 mg/L IBA,增殖系数可达4.3,并且芽苗生长健壮,为其种苗的规模化繁育提供技术基础。

Abstract: Lycopersicon esculentum ‘Xiariyangguang’is an imported high- level tomato variety recently, which gets the favor of market because of high yield and high quality. However, it is difficult to be planted in large scale for marketing because of its expensive seeds, which are all hybrid F1 generation. Base on the plant in vitro culture techniques, the seedlings ofLycopersicon esculentum‘Xiariyangguang’will be propagated rapidly. Therefore, it is a feasible way for promoting this variety in production. In order to rapidly establish aseptic lines ofLycopersicon esculentum‘Xiariyangguang’and obtain high-performance proliferation of its shootsin vitro , the key techniques were investigated by using its lateral buds as materials during itsin vitro culture. The results showed that after its explant was disinfected by using 0.1% mercuric chloride with some Tween 20 treatment for 8 min, the aseptic lines of Lycopersicon esculentum ‘Xiariyangguang’could be established in media MS 0.1 mg/L BA 0.1 mg/L IBA, whose seedling rate was 46.7%. The medium MS 0.5 mg/L BA 0.1 mg/L IBA was suitable for bud seedling subculture and proliferation to obtain healthy and strong plantlets. Its multiplication rate could reach 4.3. These techniques could lay the foundation for largescale propagation ofLycopersicon sculentum‘Xiariyangguang’seedlings.
