离体与非离体条件对豌豆根尖有丝分裂的影响

摘要: 以豌豆根尖为材料,在离体与非离体条件下分别采用低温、0.002mol/L 8-羟基喹啉、0.01g/L秋水仙素及0.01g/L秋水仙素与0.002mol/L 8-羟基喹啉混合液(V:V=1:1)进行预处理,对有丝分裂指数,中期分裂指数进行了分析。结果表明,离体条件下,0.002mol/L 8-羟基喹啉预处理的有丝分裂指数最高,比对照提高了2.15%,低温预处理的中期分裂指数最高,比对照提高了2.50%;非离体条件下,低温预处理的有丝分裂指数最高,比对照提高了2.51%,0.01g/L 秋水仙素预处理的中期分裂指数最高,比对照提高了1.59%。整体而言,豌豆有丝分裂指数和中期分裂指数是非离体条件下各种预处理均高于离体条件下各种预处理。

Abstract: The objective of this study was to detect the mitotic index and metaphase index of low temperature, 0.002mol/L 8-quinolinol, 0.01g/Lcolchicines, intermixture of 0.002 mol/L 8-quinolinol and 0.01g/L colchicines (v:v=1:1) on pea root tip cells under the condition of in vitro and nor in vitro respectively. Under the condition of in vitro, the mitotic index was 2.15% higher than that of the check when the root tip cells were treated in 0.002mol/L 8-quinolinol while the metaphase index got 2.50% higher than that of the check when cells were treated in low temperature. Under the condition of non in vitro, mitotic index of was 2.15% higher than that of the check when cells were treated in low temperature while the metaphase index reached 1.15% higher than that of the check in 0.01g/Lcolchicines. The mitotic index and metaphase index were much higher in vitro than that in non in vitro.
