灵芝金属硫蛋白基因的克隆及序列分析

摘要: 采用RT-PCR和3′-RACE相结合的方法获得了灵芝(Ganoderma lucidum)金属硫蛋白的cDNA序列。该序列全长为315bp,5′-端非翻译区为47bp,3’端非翻译区具有166bp,开放阅读框长度为102bp(包括一个终止密码子),可编码33个氨基酸。在其编码的氨基酸序列中半胱氨酸(Cys)含量最高,占21.2%,其次是丝氨基酸Ser(S)和丙氨基酸Ala(A),均占12.1%。对其编码的氨基酸序列进行分析发现该序列具有金属硫蛋白的保守序列模式,是金属硫蛋白家族的成员。

Abstract: A partial sequence of Ganoderma lucidum metallothionmerin(MT)gene was identified by RT-PCR and 3’RACE method. The full-length cNDA of G lucidum MT was of 315bp by including a 5’ untranslated region( UTR) of 47 bp,a 3’UTR of 166bp and an open reading frame (ORF) of 102 bp encoding a polypeptide of 33 amino ancids. The predicated protein from G lucidum MT gene sequence contained 7 cysteine residues organized in concerve motifs as classically described for MTs. The contens of cysteine is 21.2%, glycine and alanine were 12.1%. The result sequence analsis indicated that GlMT from Ganoderma lucidum was the member of metallothionmerin family.
