影响文心兰原球茎增殖培养之因素分析

Analysis on Factors Effecting in PLB multiplication culture of Oncidium

摘要: 以蜜糖文心兰茎尖诱导形成的原球茎(PLB)为试材,研究了影响文心兰原球茎增殖的主要因素,结果表明:采用改良1号为基本培养基,配合6-BA 2.0mg/L+NAA 0.2mg/L,添加香蕉泥50g/L+Peptone 1g/L作为有机添加剂、2%的白糖作为外源碳源,以液体振荡方式进行培养,这样的组合是原球茎增殖培养的最佳组合,能够获得最大的繁殖效率,有利于原球茎形态的保持。研究还表明,同一外植体的原球茎繁殖代数控制在9~10代即繁殖种苗数量控制在1.5万~2.0万以内,既能保证较高的增殖速度又能避免变异种苗的发生。

Abstract: The main factors of PLB multiplication culture were studied by using PLB of Onc. Sweet Sugarr which was induced from shoot-tip culture. The results showed that the best culture combination was the modified 1 basic medium supplemented with 2.0mg/L 6-BA and 0.2mg/L NAA, 50g/L banana mud and 1g/L peptone as organic additive, 2% sugar as carbon source, and liquid culture by oscillating, in which the best efficiency of reproduction was able to attain and was favorable to keep the morphology of PLBs. The test also showed that the descends of PLB which was in the same explant should control in 9~10 descends, which mean that the amounts of propagating seedlings should be restricted to 15~20 thousand, and it could assure superior multiplying rate and avoid the happening of variant seedlings.
