摘要: 摘 要:【研究目的】本文为了获得藏灵菇嗜热链球菌Tx菌株胞外多糖(EPS)的纯品,探索了EPS粗品的纯化方法;【方法】利用Sepharose CL-6B进行凝胶分级纯化,并对EPS纯品采用紫外全波长扫描、凝胶色谱鉴定及EPS纯度测定;【结果】最佳纯化条件:采用0.05~0.06 mol/L氯化钠溶液梯度洗脱,流速为0.25 mL/min,样品上样浓度为1.0 mg/mL,上样量为1.0 mL;【结论】在最佳纯化条件下得到的EPS纯度为98.01%,是纯化前的1.4倍,为今后进一步研究EPS结构与功能提供了实验基础。

Abstract: Abstract:【OBJECTIVE】In order to get the pure exopolysaccharides produced by Streptococcus thermophilus Tx from Tibetan Kefir. The purification methods were analyzed, and at last the best purification method was found;【METHOD】Sepharose CL-6B was used to make a classical purification, Using ultraviolet scan and chromatogram to identify the purification effect, the purification detection;【RESULTS】Optimization purification conditions: adoption sodium chloride 0.05~0.06 mol/L, the velocity 0.25 mL/min, the sample concentration 1.0 mg/mL, the sample capacity 1.0 mL;【CONCLUSION】On optimization purification conditions EPS purity is 98.01%, 1.4 times larger than former, which provides the basement for the study of EPS structure and function.
