IGF1对不同培养代数梅花鹿鹿茸间充质层细胞增殖的影响

Effects of Insulin-Like Growth Factor 1 on the Proliferation of Sika Deer Antler Mesenchymal Layer Cells at Different Generation

摘要:

【研究目的】通过细胞培养技术,探讨胰岛素样生长因子1(IGF1)对体外培养不同代数的梅花鹿鹿茸间充质层细胞增殖的影响。【方法】分离、培养生长90d的梅花鹿鹿茸间充质层细胞,将传代培养第2代、5代、8代细胞经含有不同浓度的IGF1(0,1,3,和10 nM)作用,培养24 h后用3H-胸腺嘧啶核苷掺入测定法检测蛋白合成放射性含量。【结果】不同浓度IGF1处理组的蛋白合成放射性含量的平均值和最高值都是离体培养2代的细胞最高(分别为32146和35973 DPM/mg);培养5代的细胞(分别为2781和3206 DPM/mg)已明显下降;第8代时(分别为489和736 DPM/mg)又进一步下降,并且与对照组无显著的差异。【结论】不同培养时期的梅花鹿鹿茸间充质层细胞对IGF1刺激的敏感度不同,取材于生长90天鹿茸的间充质层细胞在离体培养8代后已失去了对IGF1刺激的敏感性。

Abstract:

【Objective】To investigate the effects of insulin-like growth factor 1 (IGF1)on the proliferation of sika deer antler mesenchymal layer cells from different passages in vitro. 【Method】The mesenchymal layer cells were isolated from sika deer antlers of growing 90 day and cultured. The second, fifth and eighth population cells were incubated with 0, 1, 3, and 10 nM IGF1. Thymidine incorporation assay was applied after 24 hours of culture respectively to determine the radioactivity content of protein synthesis. 【Results】The average and maximal radioactivity content of protein synthesis value of the proliferation of cells from growing 90 day’s antler in the experimental group were the highest in the second population cells (32146 and 35973 DPM/mg respectively), decreased in the fifth population cells materially (2781 and 3206 DPM/mg), and decreased continuously in the eighth population cells (489 and 736 DPM/mg) and there was no significant difference compared with the control group. 【Conclusion】The sensitivity of sika deer antler mesenchymal layer cells to IGF1 varies from various passages. There was no difference in sensitivity to IGF1 in the eighth population cells at 90 days growing period of antler in vitro.
