The Study of Efficient Plant Regeneration of Solanum lycopersicum ‘Black’

Abstract: Procedures for callus induction, shoot regeneration, multiplication of shoot and rooting from regenerated shoots of ‘Black’ tomato were optimized by the study of several factor. The highest callus induction frequency was obtained from leaf explants on MS medium containing 1.0 mg/L benzylaminopurine (6-BA) with 0.1 mg/L α-naphthaleneacetic acid (NAA). The formed callus was transferred to regeneration medium supplemented with 6-BA and IBA and the best hormonal treatment to shoot regeneration was the medium with 1.5 mg/L 6-BA and 0.2 mg/L 3-indolebutyric (IBA). The medium containing 4.0 mg/L kinetin (KT) and 0.01 mg/L IBA was found to be the optimal hormonal treatment for multiplication. Also the medium with 0.05-0.08 mg/L NAA was found to be the best treatment for rooting, transplanting survival rate was 92%.
