Abstract: This paper aimed to study the determination method of spirotetramat residue in cucumber and soil by high performance liquid chromatography, and the degradation dynamics and residue rule were analyzed by this method in Hunan, Zhejiang and Guangdong in 2014. The samples were extracted by acetonitrile and detected with HPLC UV-detector. The results showed that, when the adding concentration of spirotetramat was 0.05-1.0 mg/kg in cucumber and soil, the average spiked recoveries were 95.75%-98.91% and 89.37%-99.78%, the coefficients of variation were 1.26%-2.89% and 1.62%-1.99%, and the minimum detectable concentration by this method was 0.025 mg/kg. The half-life period of spirotetramat in cucumber and soil in Hunan, Zhejiang and Guangzhou was 2.67-8.82 d and 1.95-6.19 d respectively under natural environmental condition. Spirotetramat could not be detected in cucumber and soil after 5 days of the last application with recommended dosage, and the residue quantity of spirotetramat was below the MRL value of melon and fruit (0.2 mg/kg).
