Induction of Callus and Organogenesis from Scales in Chongming Daffodil

Abstract: In this study, the scales were used as explants to induce callus and organogenesis of Chongming daffodil, in order to obtain the most favorable conditions of tissue culture. The influencing factors of induction of callus and organogenesis, such as low temperature treatment, the scales position and different combinations and concentrations of hormones were analyzed. The results showed that: the inner scales treated with low temperature (4℃) for 3 weeks was the suitable explants for inducing callus, the induction rate was 48.4%. The differentiation rate of callus was 23.3% on the MS medium with 1 mg/L BA, 0.1 mg/L 2,4-D, 30 g/L sucrose and 4 g/L agar. The experiment explored the optimal tissue culture condition to obtain the bulbsin vitro from the scales, and provided technical support for the large-scale production of Chongming daffodil.
