淡水黑鲷5种同工酶的组织特异性研究

Studies on 5 isozymes in different tissues of Hephaestus fuliginosus

摘要:

运用聚丙烯酰胺垂直梯度凝胶电泳法对淡水黑鲷(Hephaestus fuliginosus)6种组织(肝脏、心脏、脾脏、肾脏、肌肉和眼)的5种同工酶(酯酶(EST)、过氧化氢酶(CAT)、过氧化物酶(POD)、苹果酸脱氢酶(MDH)、乳酸脱氢酶(LDH))进行了研究,并对其同工酶位点及酶谱进行了分析。结果表明,淡水黑鲷5种同工酶系统具有不同程度的组织特异性。EST检测到2个位点,每个位点都有2个等位基因。CAT检测到1个基因位点,存在2个等位基因。POD酶谱较复杂,有5个位点,Pod-2,3,4,5都检测到2个等位基因。MDH具有s-MDH和m-MDH两种类型,在各组织中均表达,s-MDH只形成一种二聚体,m-MDH仅在肝脏中有2条带,其余组织中为1条带。LDH在6种组织中都表达, Ldh-5只在心脏中能检测到。

Abstract:

Five isozymes (EST, CAT, POD, MDH, LDH) in six tissues (liver, heart, spleen, kidney, muscle, eye) of Hephaestus fuliginosus were studied by using vertical polyacrylamide gel gradient electrophoresis. Zymograms and loci of these 5 isozymes were analyzed. The results showed that these five isozymes presented tissue specificity to some degrees. Two sites were detected with EST and each site was encoded by 2 loci. Only one site was detected with CAT which included 2 loci. The zymogram of POD was relatively complicated, includeding 5 sites and 4 of which had alleles. Two types of MDH (m-MDH and s-MDH) were found in six tissues: s-MDH only showed one dimer; m-MDH had 2 bands in liver and 1 bands in other tissues. LDH was detected in six tissues and Ldh-5 was only found in heart.
