孕酮人工抗原的合成及间接ElISA法鉴定

摘要:本研究采用二环己基碳二亚胺法将孕酮衍生物P4-11α-半琥珀酸酯与载体BSA和OVA进行偶联,制备了孕酮人工完全抗原(免疫原和检测原),然后免疫Balb/c小鼠,制备孕酮多克隆抗体。用紫外光谱扫描法鉴定偶联是否成功并用间接Elisa法对孕酮多克隆抗体进行鉴定;结果显示,孕酮和BSA、OVA的偶联比分别为18:1、10:1,免疫血清孕酮抗体效价高达1:25600,这说明成功制备了免疫原和检测原,为孕酮胶体金试纸条的研制奠定了基础。

Abstract:

Abstract: two kinds of artificial antigen for progesterone(P4) were synthesized and identified in this research. Progesterone-11alpha-hemisuccinate was conjugated with carrier protein Albumin Bovine V(BSA) and Albumin egg(OVA) by dicyclohexylcarbodiimide(DCC).Then,10 Balb/c mice were injected with immunogen P4-BSA to produce antibody against P4. The prepared artificial antigens were accredited by Ultra violet scan and the titer of antibody against P4 was detected by Elisa. The results show that the conjugation molecular ratios of P4 with BSA and OVA are 18:1 and 10:1 and the titer is as high as 1:25600.
