响应面法优化银翘天甘含药血清制备工艺

Optimization of Preparation Technology of Medicated Serum of Yinqiaotiangan in Mice by Response Surface Methodology

摘要:

为了优化银翘天甘含药血清的制备工艺,在单因素试验的基础上,选取采血时间、给药次数及给药浓度为自变量,以小鼠血清中绿原酸和连翘苷含量的综合评分为响应值,采用响应面分析方法,研究各自变量及其交互作用对综合评分的影响。利用Design-expert软件分析得到二次多项式回归方程的预测模型,并确定了银翘天甘含药血清最佳制备工艺条件为:采血时间30 min、给药次数4次、给药浓度0.02 g/mL,理论计算综合评分为95.62%,小鼠血清中绿原酸浓度为33.65 μg/mL,连翘苷浓度为193.86 μg/mL。该研究为银翘天甘含药血清的制备奠定了基础。

Abstract:

In order to optimize preparation technology on medicated serum of Yinqiaotiangan in mice, on the base of single factor investigation, taking-blood time, administration times, administration concentration were selected for the independent variable and comprehensive score on the concentration of chlorogenic acid and forsythia in the mice serum was as response value. Effects of every independent variable as well as their interactions on response value were studied by response surface analysis. The predictive model of two polynomial regression equations was analyzed by Design-expert software. The results showed that preparation technology was concluded as take blood time 30 min, administration times 4 times and administration concentration 0.02 g/mL. In theoretical calculation, comprehensive score was 95.62%. In this condition, the concentration of chlorogenic acid was 33.65 μg/mL and concentration of forsythia was 193.86 μg/mL in mice serum. This study provided the foundation for preparation technology on medicated serum of Yinqiaotiangan.
