鹅A-FABP基因内含子Ⅰ序列的分析研究

Sequence Analysis in the Intron-I of A-FABP Gene in Geese

摘要:

为了探讨鹅脂肪型脂肪酸结合蛋白(Adipocyte Fatty Acid-Binding Protein,A-FABP)基因内含子Ⅰ的序列长度、多态性及与其他物种的同源性。根据鹅A-FABP mRNA(AF472610)和鸡A-FABP基因组序列(AY675941)设计1对引物,扩增鹅A-FABP基因内含子Ⅰ序列;利用PCR-SSCP技术和序列比对,开展鹅A-FABP基因内含子Ⅰ的多态性研究。结果表明:鹅A-FABP基因内含子Ⅰ序列长为1473 bp,与其他12个物种序列比较,发现其与鸭的同源性达到94.42%,与鸡的同源性为81.82%。经SNPs检测发现,P2和P3扩增产物存在变异位点。遗传多态性分析表明,A、B两个等位基因的基因频率分别为0.597和0.403,多态信息含量(PIC)是0.365,基因型分布未偏离Hardy-Weinberg平衡状态(χ2=0.020,P>0.05);D、E两个等位基因的基因频率分别为0.483和0.517,PIC是0.375,基因型分布偏离Hardy-Weinberg平衡状态(χ2=6.782,0.01

Abstract:

To study the sequence length in Intron-I of adipocyte fatty acid-binding protein (A-FABP) gene, the polymorphism and homology of the other species. A pair of primers were designed to amplify the Intron-I of goose A-FABP gene, according to the sequences of A-FABP mRNA in goose (AF472610) and A-FABP gene in chicken (AY675941) and the genetic polymorphisms of Intron-I of goose A-FABP gene were investigated by PCR-SSCP and sequence alignment. The results showed that sequence size of Intron-I of goose A-FABP gene was 1473 bp, which showed the highest homology with duck (up to 94.42%) and the homology with chicken (up to 81.82%) when aligned with same gene sequence of 12 other species; Genetic polymorphism analysis by SNPs showed that the frequencies of two alleles A and B were 0.597 and 0.403 respectively, polymorphic information content (PIC) was 0.365, genotype distribution of which was in Hardy-Weinberg equilibrium (χ2=0.02, P>0.05); the frequencies of two alleles D and E were 0.483 and 0.517 respectively, PIC was 0.375, genotype distribution of which was not in Hardy-Weinberg equilibrium (χ2=6.782, 0.01
