昼夜不同温度对2种Bt棉苗期叶片杀虫蛋白表达量和棉铃虫死亡率的影响

Effects of Alternating Temperatures Day and Night on Cotton Bollworm Mortality and Insecticidal Protein Expression of Two Kinds of Bt Cottons

摘要: 为探讨Bt棉的抗虫性对不同昼夜温差变化的响应机理,应用生物测定和ELISA方法,于苗期对2种不同来源的Bt棉棉株进行连续96 h的不同昼夜温度处理,用处理后的叶片饲喂棉铃虫,研究昼夜温度变化对Bt蛋白表达及棉铃虫死亡率的影响。结果表明,白天高温35℃,夜间低温18℃,能显著影响Bt棉蛋白表达量和幼虫死亡率,且随着胁迫时间的延长,杀虫蛋白含量和幼虫死亡率急剧下降;虽然白天高温35℃,夜间正常温度28℃,也会影响杀虫蛋白表达量和幼虫死亡率,但影响程度较小,且随着胁迫时间的延长,Bt蛋白的下降幅度趋于稳定。2个不同基因来源的品种间,美国棉‘邯杂154’蛋白表达量高于‘皖棉11’,且美国棉品种受到昼夜温度变化的影响较小。白天极端高温,晚上极端低温可以显著影响Bt棉抗虫性,并且胁迫时间越长,影响越强,该研究结果将为中国棉花种植过程中经常出现的极端天气下Bt棉的抗虫性安全应用提供理论基础。

Abstract: To discuss the response mechanism of Bt cotton resistance to Helicoverpa armigera under the diurnal temperature condition, two different Bt cotton varieties were used as materials for mensurating insecticidal protein content from cotton leaves at seedling stage and investigating the death rate of cotton bollworm fed on cotton leaves. In the experiment, temperature stress (35℃/28℃ or 35℃/18℃) were carried out for 96 h in the artificial climatic chamber in order to study the influence of alternating temperatures day and night on cotton bollworm larval mortality and insecticidal protein expression. The results indicated that when day/night temperature was 35℃/18℃, it could significantly affect Cry1Ac protein content and cotton bollworm larval mortality, and in pace with the time extending, Cry1Ac protein content and larval mortality suffered a sharp decline. While 35℃/28℃ of day/night temperature also affected the insecticidal protein expression and larval mortality, but it had only small influence degree. And as the time extended, Cry1Ac protein content had a stable reduction. Between the two Bt cotton varieties from different gene sources, cotton ‘Hanza 154’ from USA had higher insecticidal protein content than ‘Wanmian11’ from Anhui. Bt cotton from USA was less affected by diurnal temperature. The extremely high temperature at daytime and the very low temperature at night might significantly affect Bt cotton resistance to cotton bollworm. What’s more, the longer the stress time, the stronger the effect degree. It will provide a theoretical basis for security applications of Bt cotton in frequently extreme weather during the process of cotton cultivation in China.
