摘要:

通过探究不同强化处理措施对铜污染土壤微生物多样性的影响,以期为解释土壤微生物影响植物吸收累积重金属的机理以及提高植物-微生物联合修复重金属污染效率提供了基础数据和参考。研究采用田间小区试验,基于高通量测序技术对不同强化处理措施下的土壤微生物进行16SrDNA测序,揭示其微生物群落组成和差异。结果表明:5种处理下微生物群落共覆盖42个门,145个纲,359个目,562个科,1060个属,2290个种。在门水平上,不同处理土壤微生物群落组成较为相似,优势物种相同,丰度较大的门类为酸杆菌门、变形菌门、放线菌门、绿弯菌门和厚壁菌门。在OTU水平上,不同处理土壤微生物群落组成具有显著差异,对微生物群落影响排序为:铜胁迫>螯合剂>有机物覆盖;不同处理出现了特异物种,空白对照和其他处理差异性最大,铜胁迫处理和有机物覆盖处理土壤微生物群落组成最为相近。铜胁迫均增加了土壤微生物群落的丰富度;螯合剂处理下土壤微生物群落的多样性和丰富度均最大,且群落多样性和有机物覆盖处理的差异达到了显著水平。

Abstract:

The effects of different intensified measures on microbial diversity in copper contaminated soil were explored to provide basic data and reference for explaining the mechanism of soil microorganisms affecting the absorption and accumulation of heavy metals by plants and improving the efficiency of plant-microbe combined remediation of heavy metal contaminated soil. Illumina-miseq high-throughput sequencing technology was used to sequence the V3-V4 region fragments of 16SrDNA gene under different intensified treatment measures in combination with field plot experiment, to reveal the composition and difference of microbial communities. The results show that a total of 42 phyla, 145 classes, 359 orders, 562 families, 1060 genera and 2290 species are identified in five different treatments. The composition of soil microbial community in different treatments is similar at phylum level, and the dominant species are same. The phyla with high abundance are Acidobacteria, Proteobacteria, Actinobacteria, Chloroflexi and Firmicutes. There are significant differences in soil microbial composition among different treatments at OTU level, and the order of effect on microbial community is copper stress > chelating agent > organic matter cover. Specific species appear in different treatments and the difference between blank control treatment and other treatments is the largest. The microbial community composition of copper stress treatment and organic matter cover treatment are the most similar. Cu stress increases the richness of soil microbial community. The diversity and richness of soil microorganisms are the highest under chelating agent treatment, and the difference of microbial community diversity between chelating agent treatment and organic matter cover treatment reaches a significant level.
