粗胫翠尺蛾视蛋白基因的克隆及灯光对其表达量的影响

Identification and Characterization of Opsin Genes from Thalassodes immissaria and Gene Expression After White LED Treatment

摘要:

灯光防控技术在国内荔枝和龙眼主要产区被推广应用,为了探究该项技术对于荔枝龙眼梢期头号害虫——尺蛾的视觉刺激响应的影响,笔者以粗胫翠尺蛾为研究材料,利用转录组测序结果和RACE技术获得2个视蛋白基因,并通过RT-qPCR技术分析2个视蛋白基因的发育模式以及在光照处理后的表达变化。结果表明,扩增得到TiLWTiUV视蛋白基因分别编码374个和378个氨基酸,其序列二级结构主要由α-螺旋和随机卷曲组成,均包含不同数量的视蛋白家族所特有的多个保守氨基酸位点。在粗胫翠尺蛾整个发育周期的不同发育阶段中,均可检测到2个视蛋白基因,但两者在不同发育阶段的表达水平有差异。在光照处理后,成虫头部TiLWTiUV的表达量均受到不同程度的影响。与对照组相比,光照处理后24 h,TiLW在雌、雄虫头部分别上升12.96倍和降低8.61倍,而TiUV在雌雄虫头部分别上升1.75倍和降低3.10倍。TiLWTiUV参与粗胫翠尺蛾在光照刺激下的应激过程,光刺激可引起粗胫翠尺蛾2个视蛋白基因表达变化。

Abstract:

Currently, lighting control technology is promoted in litchi and longan cultivation areas in China. To explore the impacts of the lighting control technology on litchi leaf looper, 2 opsin genes were cloned using RACE technology based on the transcriptome sequencing results from Thalassodes immissaria, and then expression changes of the 2 opsin genes were detected after light treatment to T. immissaria by RT-qPCR. The results showed that the open reading frames of TiLW and TiUV encoded 374 and 378 amino acids, respectively. Results of secondary structure analysis of the 2 opsin genes showed that both of them were mainly composed of α-helix and random coil and contained typical conserved domains of opsin family. The 2 opsin genes could be detected at different developmental stages of the whole development cycle, but their expression levels were different at different developmental stages. After light treatment, the expression levels of TiLW and TiUV in the head of adults were affected by different degrees. Compared with the control group, after the light treatment of 24 h, TiLW was increased by 12.96 times and decreased by 8.61 times in female and male heads, respectively, while TiUV was increased by 1.75 times and decreased by 3.10 times, respectively. The results of this study show that TiLW and TiUV are involved in the stress process of light stimulation, and light stimulation could cause changes in the expression of the two opsin genes.
