甜菊叶片β-葡萄糖苷酶的提取及其性质研究

β-glucosidase from Stevia Leaves: Extraction and Characteristics

摘要:

为研究β-葡萄糖苷酶在甜菊糖苷降解代谢中的作用,并为今后通过抑制甜菊糖苷的降解从而提高其含量的育种工作等奠定理论基础。本研究以甜菊为试验材料,首先对甜菊叶片β-葡萄糖苷酶的提取及酶活性测定方法进行了优化,并进一步对该酶性质进行了初步分析。结果表明,以pH 7.0的50 mmol/L磷酸缓冲液提取,研磨时添加等量PVP,粗酶液经20%~40%饱和度硫酸铵进行盐析的酶液其酶活性最高;以pNPG为底物测定其酶活性时37℃孵育60 min最佳。酶性质分析结果表明,该酶具有较高的热稳定性和pH值稳定性,其最适温度为30℃,最适pH 7.0;4℃保存时酶活性逐渐降低,至26天时酶活力仅余26.4%;在9种常见金属离子中Mn 2+、Fe 2+对该酶有显著的激活作用,而Cu 2+、Hg 2+对该酶的抑制作用较明显。

Abstract:

The paper aims to study the role of stevia β-glucosidase in catabolism of steviol glycosides, and to lay a theoretical foundation for breeding of increasing steviol glycosides content by inhibiting its degradation in future. Stevia rebaudiana was used as test material in this study, and we firstly optimized the methods of extraction and determination of enzyme activity of β- glucosidase from stevia leaves, and then preliminarily analyzed the enzyme properties further. The results indicated that: the enzyme solution, which was extracted by 50 mmol/L phosphate buffer with pH 7.0, and added equivalent PVP during grinding, then salted out with 20%-40% saturation ammonium sulfate by the method of ammonium sulfate grade precipitation, had the highest enzyme activity; using pNPG as the substrate to determine enzyme activity, the best method was reacted at 37℃ for 60 min; and the results of enzyme property analysis showed that: stevia β-glucosidase possessed sound thermal stability and pH stability, its optimum temperature was 30℃, the optimum pH value was 7.0; but in 4℃, the enzyme activity gradually reduced, and was only 26.4% up to 26 d; among 9 common metal ions, Mn 2+ and Fe 2+ had significant activation effect on the enzyme, while Cu 2+ and Hg 2+ had obvious inhibition effect.
