Psidium guajava L.: Genetic Diversity Analysis Based on ISSR Markers and Construction of Fingerprints

Abstract:

In order to study the genetic diversity of 59 guava germplasm resources and to map the fingerprint, DNA fingerprinting of guava was studied by ISSR technique and UPGMA cluster analysis. The 10 ISSR core primers obtained by screening were used to amplify 59 guavas samples, and a total of 126 bands were obtained, including 113 polymorphic bands, with a polymorphism rate of 89.68%. According to UPGMA clustering analysis, the genetic similarity coefficient of 59 guava germplasm resource samples ranged from 0.50 to 0.96, and the average genetic similarity coefficient was 0.73. According to the genetic distance, the 59 germplasm resources could be divided into four groups, group 1 contained 34 germplasm resources, group 2 contained 6 germplasm resources, group 3 contained 18 germplasm resources, and the sample number C26 'strawberry' guava was the fourth group alone. The DNA fingerprints of 59 guava germplasm resources were successfully constructed, which could be used for the classification and identification of guava germplasm resources.
