Abstract:

To study the effect of harvesting time on the quality of Radix Astragali in the cold area of Liupan Mountain in Ningxia, we established the HPLC fingerprint and determined the content of 5 components of Radix Astragali. The HPLC fingerprint was established by gradient elution with acetonitrile-methanol and acetic acid buffer salt in 260 nm, and the contents of calycosin-7-glucoside, formononetin, ononin and calycosin were determined with DAD detector; the content of astragaloside Ⅳ was determined with acetonitrile and water isocratic elution and ELSD detector. The fingerprint of 14 batches of Radix Astragali from different harvesting time had 15 common peaks. The contents of calycosin-7-glucoside and ononin were higher in May, and the contents of calycosin-7-glucoside and astragaloside Ⅳ were higher in October, and the total peak area of the shared fingerprint peaks was relatively high in May and October. In the cold area of Liupan Mountain in Ningxia, Radix Astragali is preferably harvested in May or October, and HPLC fingerprint combined with content determination can objectively and effectively evaluate the quality of Radix Astragali at different harvesting periods.
