Construction of SSR Fingerprint and Molecular Identity Card of Shandong Elite Julans regia Cultivars—Based on Capillary Electrophoresis Analysis

Abstract:

The study analyzed the genetic background and constructed the molecular ID cards of Shandong elite Julans regia cultivars, aiming to provide technical support for breeding elite walnut cultivars and quality traceability. In this study, 17 walnut cultivars were used as the test materials to construct the fingerprints by using fluorescent SSR markers, and the data were analyzed at the molecular level. Nei's genetic distance ranged from 0.181 to 0.720 and the genetic similarity coefficient ranged from 0.267 to 0.867 among the 17 walnut cultivars. Five pairs of SSR primers were screened to construct the fingerprints of the 17 walnut cultivars. The core primers could be divided into two groups: HF9 + HF15 and HF3 + HF9 + HF14, and each group could completely distinguish the 17 cultivars. Based on fingerprint and combined with commodity information, the molecular ID cards of the 17 walnut cultivars were established by digital coding. The genetic basis of the 17 walnut cultivars was wide, and the result showed that there was no obvious correlation between the cluster relationship and geographical location. The 5 core primers had a good polymorphism and could be used to construct walnut fingerprint. Molecular QR code could provide a basis for the identification of elite Julans regia cultivars.
