5 Polygonum tinctorium Ait. Populations: Genetic Diversity Analysis Based on ISSR Marker

Abstract:

The research aims at analyzing genetic diversity and relationship of 5 Polygonum tinctorium Ait. populations, and providing molecular references to the germplasm resources protection, exploitation and utilization of P. tinctorium Ait.. 5 P. tinctorium Ait. populations were used as materials. The genetic diversity was studied by simple inter-sequence repeat (ISSR) makers. The software Popgene 32 was used to analyze the amplification results, and the NTsys2.10e was used to construct a cluster map according to the unweighted pair-group method with arithmetic means (UPGMA). The results showed that a total of 155 loci were amplified by 23 primers, of which 148 were polymorphic loci; the Nei's genetic diversity coefficient (H) was 0.3538, Shannon's Information index (I) was 0.5234, the coefficient of genetic differentiation (Gst) was 0.4096, and the gene flow (Nm) was 0.7206. Taking the genetic distance 0.234 as a threshold, the 5 populations were divided into three groups, among which the Japanese population was different from others. The results indicate that the genetic difference among the populations of P. tinctorium Ait. is related to the geographical distance. This study could provide a theoretical basis for the genetic diversity of P. tinctorium Ait. and the protection and utilization of germplasm resources.
