Beet Germplasm Resources: Identification and Genetic Diversity Analysis by SCoT

Abstract:

To study the feasibility of SCoT molecular marker for identifying beet germplasm resources, we designed 80 SCoT primers to identify 48 beet germplasm resources, and analyzed the genetic diversity and genetic relationship of the germplasm resources. The results showed that 6 of the 80 SCoT primers, namely SCoT1, SCoT12, SCoT13, SCoT14, SCoT17, and SCoT23, amplified clear and highly polymorphic bands. Among them, SCoT1, SCoT12, SCoT14 and SCoT23 could separately identify the 48 beet germplasm resources. The identification of the 48 beet germplasm resources could be distinguished by combining SCoT13 and SCoT17. The cluster analysis results showed that 95.8% of germplasm resources were clustered into the same clade at the genetic distance of 0.15, which indicated that there was close genetic relationship among the 48 beet germplasm resources. The study provided a scientific basis for the identification of beet germplasm resources, hybrid combination parent selection and molecular marker-assisted breeding by SCoT markers.
