Abstract:

The evaluation system of polysaccharide fingerprint was established to identify the source and control the quality of Tetrastigma hemsleyanum. T. hemsleyanum from different areas were used as materials, the polysaccharide spectrum, monosaccharide content and composition of T. hemsleyanum from different areas were determined by PMP pre-column derivatization-HPLC and chemometric analysis. The polysaccharide fingerprint and its common pattern were established for different origins, and 8 monosaccharide chromatographic peaks such as glucose, rhamnose and arabinose were determined. The similarity of each batch of samples was high, ranging from 0.934 to 0.997. According to the score of principal component factor, the quality of polysaccharide from Suichang was the best, followed by that from Wenzhou. The composition of its monosaccharides was determined, and high content of mannose, glucose, galactose and arabinose were obtained, which was 9.094%, 11.451%, 56.695% and 11.030%, respectively, with galactose being the main component of the polysaccharide of T. hemsleyanum. In this study, we established the polysaccharides fingerprint of T. hemsleyanum and determined the composition and content of monosaccharides. The method is stable and reliable, and can be used for the source identification and quality control of T. hemsleyanum.
