Regeneration System of Pumpkin: Two Establishment Methods

Abstract:

To establish different regeneration systems of pumpkin, the cotyledon nodes of pumpkin seeds and two-day ovaries after pollination of ‘daguo miben’ were used as explants to study the two regeneration pathways of callus and embryoid development. The result showed that, in the process of callus development, 3% Naclo solution for 15 minutes was the best way to sterilize callus, MS+3.0 mg/L 6-BA+0.1 mg/L NAA was the best medium to induce cluster bud formation, MS+0.2 mg/L NAA was the best medium to induce single bud seedling formation. Both MS+4.0 mg/L 2,4-D+0.5 mg/L NAA+1.0 mg/L 6-BA and MS+0.06 mg/L TDZ could effectively induce the formation of pumpkin embryoid, and MS medium was the most helpful for embryoid seedling formation. In this study, the two developmental pathways (callus and embryoid) were successfully established for regeneration systems of pumpkin.
