Primary Study of Spores Germination of AM Fungi and the Dual Culture
with Clover Roots

Abstract: The germination ratio of spores was compared after stored different time in cool temperature and with different progenitive form. The dual culture of Gigaspora margarita spores and clover roots was done as well. The results showed: (1) The native spores stored at 4℃ for 10-50 days significantly gained higher germination ratio. The highest ratio was 30.34% in the track of 20 days store. (2) Different progenitive form had different germination ratio. In this experiment, the order was Gl. sp sporacarps>Gi. margarita>Gl. mosseae>Gl. sp spores;(3) In dual culture, the effect of host plant roots on spore’s germination was not significant, but obviously promoted branch of hypha. The groveling infection was observed. Under runner hypha which have stopped growing branched separated ones, and further developed branched absorbing structure. After one month, there were (45.76±5) spores per millimeter root. The hypha growth direction was not influenced by roots.
