Screening and Identification of Strains 1-21 degradation Polyvinyl Alcohol

Abstract: Abstract:Due to the biological method degradation of polyvinyl alcohol (hereinafter referred to as PVA) can significantly reduce processing polyvinyl alcohol wastewater cost,therefore,screening efficient PVA degradation bacteria to PVA wastewater treatment is very important. The collected soil samples after 85℃ boiling water bath were cultured in suspension,and by boric acid and I2-KI staining,and by the shake flask fermentation supernatant rescreening PVA degrading enzyme activity determination of 9 strains,selecting PVA degrading enzyme of high activity 4 strains,the highest activity of which is named 1-21,Microbiological classification and identification methods based on the strain of the morphological,physiological and biochemical characterization and 16S rDNA sequences build phylogenetic trees,strain is identified as a Bacillus amyloliquefaciens.The similarity of the 16S rDNA sequences between strain 1-21 and the type strain BCRC 11601 of Bacillus amyloliquefaciens was close to 99.77%,With biological methods to deal with PVA wastewater provide advantage strain.
