玉米SSR分子标记技术操作流程研究进展

Review on Operation Procedures of Maize SSR Molecular Marker Technology

摘要:

SSR分子标记是一种基于PCR技术的分子标记技术,以其多态性高、共显性遗传、检测简单等优点在玉米品种鉴定、杂种优势类群划分等方面有广泛的应用。DNA提取、PCR扩增、产物检测是玉米SSR分子标记的技术流程的重要环节。尽管出现了一些能够整合SSR标记流程几个环节甚至是整个流程的实验设备,但这样的仪器常常本身价格昂贵,实验药品、耗材等成本也很高。因此,近年来,科研人员对玉米SSR分子标记技术流程各步骤进行大量的优化和改进,使实验过程更加简化、高效。为此综述了玉米SSR分子标记技术流程的DNA提取、PCR扩增、产物检测3个关键环节,分析认为这一技术流程的发展体现了实验试剂集成化、实验操作标准化、实验流程自动化的总体趋势。

Abstract:

SSR is a kind of PCR based molecular marker with high genetic polymorphism, codominant inheritance, rich content and easy-detection. Maize SSR technology is widely used in varieties identification, heterosis groups establishment, and so on. DNA extraction, PCR amplification and product testing are the key links of maize SSR molecular marker technology process. Despite of the appearance of some instruments that can integrate some links or even the entire process of the SSR operation procedure, the high price of them and expensive consumables cost inhibit their application .Therefore, in recent years, a lot of optimization and improvement have been put into effect to make the procedure more simplified and efficient. In this paper, three basic circles of maize SSR molecular marker technology are summarized, including DNA extraction, PCR amplification, product testing. The tendency of the experimental reagent integration, experiment manipulation standardization, the experimental process automation is shown according to analysis.
