植物内源激素预处理方法与检测技术的研究进展

摘要:

植物激素是植物体内合成的一系列痕量有机化合物,其对植物生长发育具有重要的调控作用,但含量极低,内源激素的测定技术对研究内源激素在植物生命活动的调节作用中具有非常重要的意义。此文对主要激素的预处理方法及相应的检测方法包括生物测定法、理化测定法和免疫测定法的研究进展进行了综述,并对植物内源激素检测技术研究中存在的问题和发展前景进行了简要讨论。气相色谱法—质谱联用和高效液相色谱法是目前内源激素精确测定的主要方法,但其对前处理要求较高。而液相色谱法—质谱联用技术将高效液相色谱法对复杂样品的高分离能力,与质谱法具有的高选择性、高灵敏度的优点结合起来,成为目前广泛应用的植物激素测定方法。

Abstract:

Phytohormones were a series of trace organic compounds synthesized in plants, which played an important role in regulation on plant growth and development, but occured in extremely low concentrations. The endogenous hormones determination technology was very important tool to study their regulatory activity in the plant life. There had been a great deal of discussion on the pretreatment and determination methods with regard to main plant hormones, including biological assay, physical and chemical assay and immunoassay in this paper, as well as the challenges and prospects in the determination methods of plant hormones had been discussed. Gas chromatography-mass spectroscopy and High Performance Liquid Chromatography detection methods were currently the mainstream methods of precise determination, but which needed higher pretreatment method. Liquid Chromatography-tandem mass spectometry had emerged as an effective method for hormones quantification because it combined efficient separation in Liquid Chromatography with selectivity and specificity in MS/MS.
