Mutagenesis Application to Saccharum officinarum Shoot Tip Culture with EMS

Abstract: In order to find out the optimum dose and time period of ethyl methyl sulphonate (EMS), with which the chemical mutagenesis was applied in sugarcane by treating the stem tip meristem, variations in sugarcane cultivars (‘ROC22’ and ‘YT93-159’) were induced through in vitro mutagenesis with varying doses (24, 48, 72 μmol/L) and time periods (exposed for 2 or 4 h) of EMS. The results showed that the proliferation rate, the percentage of surviving explants, the percentage of shoot tips regenerating shoot, and regenerated shoot development and numbers of the two cultivars declined with the increasing concentration and extending time periods of EMS. Based on some factors such as the percentage of survival and the proliferation rate as well as the reduction of regeneration capacity, 48 μmol/L for 2 h or 24 μmol/L for 4h treatment in ‘ROC22’, and 72 μmol/L for 2 h or 48 μmol/L for 4 h treatment in ‘YT93-159’ were the optimum.
