Pollen viability and stigma receptivity study of Phalaenopsis

Abstract: The high activities of pollen and stigma receptivity are the keys to ensure the success of crossbreeding work. To obtain the most applicable time of artificial pollination, scanning electron microscopy and TTC (2,3,5-triphenyltetrazolium chloride) staining were conducted to study the pollen viability of Phalaenopsis ‘Tianxiang’. Meanwhile, the stigma receptivity was determined by the benzidine- H2O2 method. Results indicated that with the anthesis extending, the Phalaenopsis pollinia volume decreased, the color deepened, texture became harder and vitality became weaker; TTC tests showed that the pollen viability rate (dyeing rate) was: open 1 d <bud <buds unfold period. Stigma tests clarified that when cross-fertilizing within 10-30 d after flowering, success rate of artificial hybridization could be improved, and the rate could reach the maximum within 10-15 d after flowering.
