Screening of a Strain Producing Alkaline Xylanase and Optimization of Fermentation Conditions

Abstract: Alkaline xylanase widely used in paper making,textile and other fields.In screening the strains of the high-yield alkaline xylanase, a alkaline xylanase generating strain YH158 is isolated from the soil of the botanical garden in Jincheng City, Shanxi Province by congo red color reaction. Morphological test and 16S rDNA sequence analysis were employed to confirm as bacillus methylotrophicus. After one factor was used to optimize, and the orthogonal test including four factors and three levels were employed to analyze, the superior fermentation medium formulation was determined:wheat bran5.0%, soyabean extraction 0.7%, MnSO4 0.1%,The optimal culture condition: initial cultural pH 8.0;temperature 37 ℃ ;agitation 180 r/min; fermentation period 36 h. The enzyme activity in supernatant of the fermentation broth was 28.18 IU/mL. Xylanase produced by this bacterium shows preferred alkaline adaptability, and its hydrolysis products are rich in xylo-oligosaccharide with 2-5 xylose molecules, displaying higher industrialization prospect.
