Application Evaluation of a Modified McMaster Egg Counting Plate

Abstract:

To evaluate the performance of modified McMaster egg counting plate developed in our lab, fecal samples of rabbits and oocyst suspension of Eimeria tenella were examined and counted using both classic and modified McMaster egg counting plates. The results of floating methods with saturated sodium chloride solution showed that the numbers of oocysts in rabbit fecal samples (2-5 g) and oocyst suspension were precisely counted using the modified McMaster plate. The coincidence rate with the results of classic McMaster plate was 95.59%-99.49%. In the precipitation experiment with tap water, the oocyst numbers in rabbit fecal samples (0.3-0.5 g) and oocyst suspension were also precisely counted using the modified McMaster plate. These findings suggest that the modified McMaster egg counting plate could be used to count the oocyst or egg number in fecal specimen and suspensions in labs, and is more powerful in counting functions than the classic McMaster plate.
