Improvement of Paraffin Section Methods and Structural Observation of Apple Anther

Abstract: Paraffin section, the most common method in conventional sectioning, is often used to observe the cellular structure. This study used the apple anther as experimental material. In this study, the dehydration, dyeing and other slice making steps were modified. The extra gradients of ethanol concentration were added and the samples were stained with Ehrlich hematoxylin for 15 min. The dyeing of sample slices was more homogeneous and more complete cell structures were obtained. The results showed that, the modified paraffin section method was more suitable for the anther of apple.
