Cluster Analysis of Sugar Beet Resistance to Cercospora Leaf Spot with SRAP Makers

Abstract: The aims are to identify and analyze domestic and foreign sugar beet varieties with better resistance to Cercospora leaf spot, and provide a basis for screening and breeding excellent disease-resistant varieties in the future. Field investigation and molecular marker were used in this study. 12 pairs SRAP molecule markers were used to detect 25 domestic and foreign varieties. A total of 104 unambiguous bands were obtained by SRAP-PCR, 67 of which were polymorphic. The average ratio of polymorphic bands was 64.4%. The genetic distance was 0.4670, and the genetic similarity was 0.6269. The results of field investigation showed that nine varieties belonged to the level 2 Cercospora leaf spot disease, and three varieties had Cercospora leaf spot disease level lower than level 2. The cluster analysis showed that 25 genotypes were divided into three cluster groups based on cluster analysis (at intercept of 0.20), and the first group was divided into two sub-groups. The clustering results were consistent with the field survey results. The results showed that‘TY305’(domestic), ‘TY309’(domestic) and‘w5-1’(domestic) had better resistance to Cercospora leaf spot. SRAP molecular markers had reference value for dividing Cercospora leaf spot resistant varieties of sugar beet.
