The Pathogen Causing Root Rot in Zanthoxylum bungeanum in Mao County of Sichuan Province: Isolation and Identification

Abstract:

To identify the pathogen species causing root rot in Zanthoxylum bungeanum in Mao County, Aba Tibetan and Qiang Autonomous Prefecture, and provide theoretical reference for scientific and effective treatment of this disease, the rot roots of Z. bungeanum were collected from Mao County and the pathogen was isolated and identified by tissue isolation method, pathogenicity determination, morphological characteristics and molecular identification which was based on ITS and tef1 gene sequences. The results showed that the mycelium of pathogen was gray white at the beginning, and then became blue brown or purple red. Large conidia of the pathogen were sickle-shaped and small conidia were kidney-shaped. Phylogenetic analysis based on ITS and tef1 gene sequences showed that the pathogen was attributed to the same branch with Fusarium solani in the phylogenetic tree. According to morphology and molecular identification, the pathogen of root rot in Z. bungeanum in Mao County of Sichuan Province was identified as F. solani.
