Effect of Subculture Cycle on Kinetics of Suspension Cultures of Protocorm-like
Bodies from Dendrobium huoshanense

Abstract: To solve the problem of low growth rate and metabolism level in suspension cultures of protocorm-like bodies from Dendrobium huoshanense. Protocorm-like bodies on different periods (20, 30, 40, 50d) were cultured individually. The uptake of carbohydrate, nitrogen and phosphate from the medium and intracellular reductive sugar, nitrogen and phosphate were investigated. The growth rate of protocorm-like bodies and the accumulation of polysaccharide were analyzed too during the whole growth period. Cell age of 20~40 days was suitable for the cell cultures because favorable growth and the accumulation of polysaccharide were maintained. The cell age of 30 days was the best,which uptook carbohydrate, nitrate and phosphate faster than that of the others. The specific growth rate was the biggest and it was 0.043. The biomass and production of polysaccharides were the highest on day 36, reaching 470.0 g/L and 1.92 g/L, respectively. The cell age of 30 days was beneficial to the cell growth and the accumulation of polysaccharides.
